研究目的:

通过将自固化磷酸钙(CPC)与米诺环素软膏及医用胶原海绵组合,形成新型抗菌药物缓释骨替代材料,植入新鲜拔牙窝,观察复合材料支架消除拔牙窝炎症、支撑牙槽骨壁形态、促进骨愈合与骨量保存的临床效果,最终实现早期或即刻种植体植入修复缺牙。  

Objectives of Study:

Calcium phosphate cement (CPC) can self-set to form osteoconductive bone substitute and can also act as carrier to deliver drugs due to its non-exothermic setting reaction and dissolution properties. In this study, CPC is compounded with minocycline ointment (MINO), a tetracycline antibiotic that is effective against most of the bacteria microorganisms related to periodontal disease and benefit for bone healing in vitro and in vivo. The hypothesis is that the composite material of CPC-MINO and collagen sponge could form porous bone substitute suitable for bone formation, and could be used for alveolar ridge preservation after tooth extraction.

药物成份或治疗方案详述:

采用上海瑞邦生物材料有限公司生产的自固化磷酸钙人工骨(CPC),无菌环境下,按照使用说明,将单瓶包装的3.8g CPC粉末与1.5ml固化液混合调匀,再加入日本大阪Sunstar公司生产的盐酸米诺环素软膏(商品名:派丽奥 PERIOCLINE)两支共1.0g(含米诺环素20mg),混合调匀制成CPC-米诺环素混合糊剂;修剪无锡贝迪生物工程股份有限公司生产的50*25*2mm医用胶原蛋白海绵块,制成约1 * 1 * 5mm的胶原颗粒,混入CPC-米诺环素糊剂,填入直径8mm深度5mm的圆形PTFE模具中结固10分钟,制成CPC为基础的多孔复合材料支架。 

Description for medicine or protocol of treatment in detail:

A moldable paste of CPC (Calcium Phosphate Cement, Rebone, Shanghai, China) is prepared by mixing 3.8g CPC powder with 1.5ml setting liquid in accordance with the manufacturers instructions, and then mixing with 1.0g minocycline ointment (Periocline, Sunstar, Osaka, Japan) that contains 20mg of minocycline. Porous scaffolds of CPC composites are then made by blending CPC paste with 1*1*5mm3 collagen granules (prepared from two sheets of 25*25*3mm3 collagen sponges), and seting the composites in round PTFE molds of 8mm diameter and 5mm depth. The scaffolds were preserved in sealed aseptic bottles in refrigerator for later use.

(2)患有高血压、肝肾疾病等未能得到有效治疗。
(3)患有与骨代谢有关的任何全身性疾病。

Exclusion criteria:

(1) Uncontrolled diabetic diseases;
(2) Uncontrolled hypertension, or hepatic and renal disorders;
(3) Any systemic diseases associated with bone metabolism.
