阶段一 探索肺癌组织和正常肺组织之间微生态特征差异,寻找特征性菌群,初步评价其诊断价值; 阶段二 构建包括微生态特征的诊断预测模型,验证该模型在基于穿刺组织的肺癌诊断中的效能。  

Objectives of Study:

Stage 1: explore the difference of micro-ecological characteristics between lung cancer tissue and normal lung tissue, search for characteristic flora, and preliminarily evaluate its diagnostic value; Stage 2: Construct a diagnostic prediction model including micro-ecological characteristics, and verify the effectiveness of this model in lung cancer diagnosis based on puncture tissue.

药物成份或治疗方案详述:

利用二代测序平台行16S rDNA检测,探索性分析并比较2组间(肺癌原发灶vs. 健康肺组织)微生态特征差异。通过使用随机森林法识别关键特征菌群,进行五次交叉验证以及随机森林分析,计算重要性评分以评估变量的重要性。绘制ROC曲线评价其诊断效能。 

Description for medicine or protocol of treatment in detail:

16S rDNA test was performed using the second-generation sequencing platform. Exploratory analysis and comparison were conducted between the two groups (primary lung cancer vs.Differences in microecological characteristics in healthy lung tissues.The importance score was calculated to assess the importance of variables by identifying key feature flora using the random forest method, conducting five cross-validation and random forest analysis.ROC curve was drawn to evaluate its diagnostic effectiveness. 

纳入标准:

Inclusion criteria

排除标准:

1)术后病理证实为小细胞肺癌;
2)合并COPD、哮喘、肺纤维化等肺部慢性疾病;
3)近1个月发生急性呼吸系统感染性疾病;
4)合并口腔及牙周疾病;
5)新辅助治疗史;
6)近3个月抗生素及全身性糖皮质激素使用;
7)其他系统恶性肿瘤病史;
8)肺部手术史;
9)合并有心血管、肝脏、肾脏、造血系统等严重疾病的患者;
10)多原发性肺恶性肿瘤。

Exclusion criteria:

1) Postoperative pathology confirmed small cell lung cancer;
2) Combined with COPD, asthma, pulmonary fibrosis and other chronic pulmonary diseases;
3) Acute respiratory infectious diseases occurred in the past 1 month;
4) Combined with oral and periodontal diseases;
5) History of neoadjuvant therapy;
6) Use of antibiotics and systemic glucocorticoids in recent 3 months;
7) History of other systematic malignant tumors;
8) History of lung surgery;
9) Patients with cardiovascular, liver, kidney, hematopoietic system and other serious diseases;
10) Multiple primary pulmonary malignant tumors.
