研究目的：

揭示膝关节炎Tuina治疗对脑皮质和皮层下结构的潜在形态改变、脑白质完整性、脑功能区活动性和脑区连接的影响，为推拿在膝关节炎治疗中的临床应用提供科学依据。

Objectives of Study：

To reveal the influence of Tuina treatment on the potential morphological changes of cortical and subcortical brain structures, the white matter integrity and the functional activities and connectivity of brain regions of patients with KOA, which might provide scientific evidence for the clinical application of Tuina in management of KOA.

纳入标准：

Inclusion criteria

排除标准：

1. 在过去1个月内服用可能影响脑部成像结果的止痛药或麻醉剂；
2. 在过去一个月内接受过其他治疗；
3. 是孕妇还是哺乳期妇女；
4. 患有精神疾病、神经系统疾病、传染病、胃肠道疾病、心脑血管疾病、免疫系统疾病、呼吸系统疾病或肾脏疾病等慢性疼痛症状，或有脑损伤并丧失意识的病史；
5. 被诊断为癌症、肺结核、风湿病或类风湿性关节炎、痛风、关节外伤；
6. 有幽闭恐惧症、心脏起搏器、除颤器、心脏支架、宫内节育器等MRI禁忌症；
7. 膝关节周围有皮肤损伤。

Exclusion criteria：

1. Patients who have taken analgesics or anesthetics that may affect the results of brain imaging in the past month;
2. Patients who have received other treatment in the past month;
3. Pregnant women or lactating women;
4. Patients with chronic pain symptoms such as mental disease, nervous system disease, infectious disease, gastrointestinal disease, cardiovascular disease, immune system disease, respiratory system disease or kidney disease, or history of brain injury and loss of consciousness;
5. Patients diagnosed with cancer, tuberculosis, rheumatism or rheumatoid arthritis, gout and joint trauma;
6. Patients with claustrophobia, pacemaker, defibrillator, cardiac stent, intrauterine device and other contraindications of MRI;
7. Patients with skin injury around the knee joint.

符合条件的患者将按1:1的比例随机分为推拿组和教育组，每组30例，由负责受试者数据统计的专业人员进行简单随机分组。随机数列表将由随机数生成器生成（IBM SPSS Statistics version 21.0 software；IBM Corp.，Armonk，NY，USA）生成，并由独立助理用顺序编号、不透明和密封的信封发送给治疗师。治疗师将打开随机分配信封，并相应地将参与者分配到推拿组或教育组。

Eligible patients will be randomized in a ratio of 1:1 to the Tuina group and the Education group with 30 patients in each group using simple randomization by the professional responsible for data statistics of the subject. The random number lists will be created will be generated by a random number

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：	原始的数据记录及病例记录表，纸质版由实验负责人保存；电子版采用EXCEL及SPSS格式进行保存。
