研究目的：

客观评价益生菌混合物对孤独症儿童核心症状以及胃肠道症状和睡眠的影响，并探讨对肠道菌群以及代谢和免疫的影响。  

Objectives of Study：

The main purpose of this study are to evaluate efficacy of probiotics mixtures on the core deficits as well as gastrointestinal symptoms and sleeping problem of children with ASD. and the changes of intestinal flora abundance before and after probiotics supplementation. The potential changes of intestinal flora abundance as well as metabolomics profiles and immunity will be evaluated to investigate the potential mechanisms.

药物成份或治疗方案详述：

治疗方案： 研究人员的目标是招募160名志愿参加的孤独症患者。在开始数据收集之前，患儿监护人需要签订将给出一份详细的知情同意书。参与者在进行CARS量表评估后，将其分为轻度、中度和重度三个等级，每个等级按照独立的随机数字表进行分组，分为安慰剂组和益生菌组。两组患儿接受相同的教育和康复训练，此外，益生菌组给予益生菌混合物用温水或牛奶口服，早晚各一次，连续3个月，安慰剂组用麦芽糊精代替。所有参与者需要在基线时、1月、2月、3月和停药后3月进行量表评估并留取标本进行相关检测。研究期间需要避免服用抗生素。如有任何不良反应或疾病，应立即通知调查人员。 益生菌混合物配方： Lactobacillus, Bifidobacterium, Streptococcus 

Description for medicine or protocol of treatment in detail：

Study protocol: This is a stratified, randomized, double-blind placebo control study, with two parallel arms: placebo group and probiotic group. This study will enroll 160 ASD children. All the children will be evaluated with CAS scale and divided as mild, moderate and severe degree of ASD. A detailed informed consent will be signed by the parents or guardians before the study. All the children will be treated equally with standard education and rehabilitation program, except the treatment group will be given probiotic mixture, twice a day for 3 months, and control group will be given maltodextrin. All the children will be evaluated systematically before, and at 1, 2, 3 months as well as after 3 months treatment, which include all the forms and laboratory analysis. Antibiotics should avoid during the study, and any uncomfortable feeling and disease need report immediately. Contents of probiotics: Lactobacillus, Bifidobacterium, Streptococcus 

纳入标准：

Inclusion criteria

排除标准：

（1）三个月内服用抗生素及抗真菌药；
（2）特殊饮食：如无麸质，酪蛋白饮食，或特殊的碳水化合物饮食；
（3）三个月内服用其它益生元，益生菌，抗氧化剂；
（4）腹泻或一周内有发热等明显炎症反应；
（5）有其他神经系统症状如癫痫等；
（6）克罗恩病，炎症性肠病，食物不耐受等胃肠道疾病；
（7）肝脏疾病；
（8）1型糖尿病 ；
（9）Rett综合征(RTT)等遗传代谢性疾病的明确诊断。

Exclusion criteria：

1. Patients who took antibiotics and antifungal drugs within three months;
2. Patients with special diet: such as no gluten, casein diet, or special carbohydrate diet;
3. Patients who take other prebiotics, probiotics and antioxidants within three months;
4. Patients with diarrhea or fever within one week;
5. Patients with other neurological symptoms such as epilepsy;
6. Crohn's disease, inflammatory bowel disease, food intolerance and other gastrointestinal diseases;
7. Patients with liver diseases;
8. Patients with type 1 diabetes mellitus;
9. Definite diagnosis of Rett syndrome (RTT) and other genetic metabolic diseases.
