研究目的:

1 分析ASD儿童在执行实验任务时脑区含氧血红蛋白激活量的变化,比较分析ASD各年龄段儿童在行为实验功能上是否存在差异及脑区的变化情况,为自闭谱系障碍进行诊断提供客观依据,也对自闭谱系障碍的严重程度、康复疗效的评定、预后进行客观分级提供标准。 2 分析各组儿童执行不同任务时平均反应时间与ASD的相关行性;研究分析疾病、年龄对重复刻板行为、模仿行为的影响,并建立回归方程。 3 探索ASD群体脑网络结构与正常发展儿童在同样实验任务条件下是否存在显著的差别,分析ASD的脑网络连接强度和网络效率是否低于正常儿童,以探讨ASD儿童发病机制的神经生理基础。  

Objectives of Study:

1. To analyze the changes of the amount of oxyhemoglobin activation in the brain area of ASD children when they perform the experimental tasks, to compare and analyze whether there are differences in the behavioral experimental functions of ASD children of all ages and the changes in the brain area, so as to provide an objective basis for the diagnosis of ASD, as well as an objective grading standard for the severity of ASD, the evaluation of rehabilitation effect and the prognosis. 2. To analyze the correlation between the average response time and ASD in different tasks of children in each group; to study and analyze the influence of disease and age on repeated stereotypical behavior and imitation behavior, and to establish regression equation. 3. To explore whether there is significant difference between ASD group brain network structure and normal development children under the same experimental task conditions, and to analyze whether ASD brain network connection strength and network efficiency are lower than normal children, so as to explore the neurophysiological basis of the pathogenesis of ASD children.

药物成份或治疗方案详述:

重复刻板行为实验全过程由一名主试,主试确保儿童理解实验规则即手指动作模仿任务后,进行实验练习部分。练习后确保儿童完全理解任务内容,开始正式实验,运动指令以声音的形式呈现,被试听到声音“1”时,一方开始执行任务(完成一段简单敲击动作,不超过5声),另一方模仿任务;听到声音“2”时,角色互换,模仿者变为动作引导者,开始执行任务;听到声音“嘟”时,停止动作,全身放松休息。声音随机呈现,由主试(医生)和被试完成任务,只需采集被试的近红外数据即可。实验采用block设计,共20个block,其中10个block是模仿者-引导者(被试)任务,10个block是模仿者(被试)-引导者任务,随机呈现。每个block20s,随后休息20s,共60s。实验时间大约15分钟。利用统计学多因素回归模型对被试数据信息进行校正与控制。 模仿行为实验共有四个模仿行为任务:工具行为模仿、目的意图模仿、过度模仿、同步模仿。被试在做实验任务的同时佩戴近红外设备,在实验的过程中使用摄像机拍摄被试的模仿行为,以便后期记录分析。 

Description for medicine or protocol of treatment in detail:

The whole process of the repetitive behavior experiment is conducted by one experimenter. The experimenter make sure that the children understood the experimental rules, i.e. the finger movement imitation task, and then carried out the experimental exercise part. After the practice, make sure that the children fully understand the task content, and start the formal experiment. The movement instruction is presented in the form of sound. When the subject hears the sound "1", one side begins to perform the task (complete a simple percussion movement, no more than 5 sounds), and the other side imitates the task. When the voice "2" is heard, the roles are reversed and the imitator becomes the action leader and begins to perform the task. When you hear the sound "Du", stop the movement and relax your body. The sound is presented randomly, and the task is completed by the subject (the doctor) and the subject. Only the near-infrared data of the subject is collected. The experiment adopts block design, with a total of 20 blocks, of which 10 blocks are imitation-facilitator (subject) tasks and 10 blocks are imitation-facilitator tasks, which are presented randomly. Each block20s, and then take another 20s break, for a total of 60s. The experiment lasts about 15 minutes. Statistical multivariate regression model is used to correct and control the data information of the subjects. There are four tasks in the imitation behavior experiment: tool behavior imitation, purpose and intention imitation, excessive imitation and synchronous imitation. The subjects wear near-infrared devices while doing experimental tasks. During the experiment, cameras are used to film the subjects' imitation behaviors for later recording and analysis.

排除标准:

(1)严重失语或认知功能障碍或精神疾患等原因导致无法理解研究者指令,无法配合研究的受试者;
(2)临床痴呆评定量表(CDR)> 0.5分;
(3)有癫痫病史受试者;
(4)失用症患者;
(5)失认症患者;
(6)伴有严重心肺疾病、病情较重、生命体征不平稳等增加受试者风险的相关疾病;
(7)正在参与其他干预性研究。

Exclusion criteria:

1. Subjects who are unable to understand the researcher's instructions and cooperate with the study due to severe aphasia, cognitive dysfunction or mental illness;
2. Patients with clinical dementia rating scale (CDR) > 0.5;
3. Subjects with history of epilepsy;
4. Patients with apraxia;
5. Patients with agnosia;
6. Related diseases with serious cardiopulmonary disease, severe illness, unstable vital signs, etc. that increase the risk of subjects;
7. Patients who are participating in other intervention studies.

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

During the clinical trial, data collection is conducted according to the CRF filling of each subject. After the trial, clinical data are summarized, data cleaning, database locking and statistical analysis are conducted.
