研究目的：

通过一项前瞻、随机、并有常规标准治疗作为对照的队列研究，评估自体脂肪源性干细胞/基质细胞治疗2型糖尿病的有效性与安全性，为临床治疗2型糖尿病探索新的治疗方案。  

Objectives of Study：

This prospective, randomized parallel controlled trial will explore the safety and clinical efficacy of autologous adipose-derived stem/stromal Cells (ADSCs) versus conventional standard therapy in the treatment of type 2 diabetes mellitus and to explore new strategy for type 2 diabetes mellitus.

药物成份或治疗方案详述：

经纳入标准与排除标准筛选后，将收集的受试者按随机、平行对照试验设计的原则按照1:1的比例分为试验组与对照组，干细胞试验组50例，对照组50例。对照组给予常规标准治疗；试验组给予常规标准治疗 +自体脂肪干细胞静脉注射。 ADSC细胞治疗方案：干细胞试验组病人，将2×10^6/kg体重的ADSC细胞悬浮于生理盐水中静脉注射。 

Description for medicine or protocol of treatment in detail：

After screening with inclusion criteria and exclusion criteria, the subjects were divided into the experimental group and the control group according to the principle of randomized and parallel controlled trial design with 50 patients in the experimental group and 50 in the control group. The control group patients were given conventional standard therapy; the experimental group was given conventional standard therapy and i.v. injection of autologous adipose-derived stem/stromal cells. ADSC cell therapy protocol: ADSC (2×10^6 per kilogram of body weight) suspended in normal saline. The control group patients only received conventional standard therapy.  

纳入标准：

Inclusion criteria

排除标准：

1. 妊娠、哺乳期或近半年内有孕育打算的女性；
2. HIV、肝炎病毒、梅毒阳性；
3. 有未控制或难以控制的心脑血管、肝、肾、肺、内分泌等系统疾病，预期寿命不足1岁；
4. 有恶性肿瘤病史；
5. 有急慢性感染；
6. 1型糖尿病、妊娠糖尿病或其他特殊类型糖尿病患者；
7. 合并自身免疫性疾病；
8. 受试者在研究开始前3年内，有酗酒、药物滥用或患有精神疾病；
9. 患者在研究前3个月内参与另一项临床研究；
10. 出血倾向严重，凝血功能差(PTA<35%), 有出凝血功能障碍；
11. 研究者认为不适合纳入研究的患者。

Exclusion criteria：

1. Pregnant or considering pregnancy in the next 6-month, lactating.
2. Positive tests for HIV, HCV, HBV and syphilis.
3. Uncontrollable or difficult to control cardiovascular and cerebrovascular, liver, kidney, lung, endocrine and other systematic diseases with a life expectancy of less than 1 year.
4. History of malignancy.
5. Acute or chronic infections;
6. Gestational diabetes.
7. Known autoimmune disease.
8. Subjects had alcoholism, drug abuse or mental illness within 3 years prior to this study.
9. Patient participating in another clinical study within 3 months of this study.
10. Serious bleeding tendency, poor coagulation function (PTA <35%).
11. The investigator realizes the patient is unsuitable for the study.
