研究目的:

本研究旨在解决GLP-1受体激动剂类药物减重人群停药后的体重反弹难题。通过评估“高蛋白饮食”与“小剂量药物维持”及其联合方案抑制体重反弹的干预效果,同时检测肠道微生物群及其相关代谢物深入解析其在体重反弹中的作用机制,旨在揭示微生态重塑是否能够削弱减重后易反弹的“肥胖记忆”效应,为制定预防反弹的长效体重管理策略提供科学依据。  

Objectives of Study:

This study aims to address the challenge of weight rebound following discontinuation of GLP-1 receptor agonists in individuals achieving weight loss. By evaluating the efficacy of a high-protein diet, low-dose maintenance of the medication, and their combination in suppressing weight rebound, while simultaneously examining gut microbiota and related metabolites to elucidate their mechanistic roles in weight rebound, this study seeks to determine whether gut microbiota remodeling can attenuate the 'obesity memory' effect that predisposes individuals to rapid rebound after weight loss, thereby providing a scientific basis for developing long-term weight management strategies to prevent rebound.

纳入标准:

Inclusion criteria

排除标准:

1. 排除有甲状腺髓样癌史或家族史、2型多发性内分泌肿瘤综合征患者;排除有胰腺炎病史者; 2. 排除严重肝功能损害或重度肾功能损害(eGFR<30 mL/min/1.73 m^2)受试者; 3. 特殊状态:排除妊娠或哺乳期妇女; 4. 患有严重器质性疾病、恶性肿瘤性疾病或正在进行放化疗及免疫治疗的患者; 5. 酗酒者(1周饮酒5次以上,平均每次2两白酒或半斤黄酒或5瓶啤酒以上); 6. 过去3个月内使用过抗生素,并持续3天或以上者; 7. 肢体残疾、有严重的精神疾患,或其他原因引起的生活不能自理、不能清楚地回忆、回答问题者; 8. 进行过胃肠道手术(包括减重手术)者; 9. 不能保证充足的时间参与本项目的。

Exclusion criteria:

1. Patients with a personal or family history of medullary thyroid carcinoma or multiple endocrine neoplasia type 2; exclude those with a history of pancreatitis. 2. Subjects with severe hepatic impairment or severe renal impairment (eGFR < 30 mL/min/1.73 m^2). 3. Special conditions: pregnant or lactating women. 4. Patients with severe organic diseases, malignant neoplastic diseases, or those undergoing radiotherapy, chemotherapy, or immunotherapy. 5. Individuals with alcohol use disorder (alcohol consumption > 5 times per week, averaging > 100 mL of liquor, > 250 mL of rice wine, or > 5 bottles of beer per occasion). 6. Those who have used antibiotics for 3 consecutive days or longer within the past 3 months. 7. Individuals with physical disabilities, severe mental disorders, or other conditions leading to inability to care for themselves, or inability to recall clearly and answer questions. 8. Those who have undergone gastrointestinal surgery (including bariatric surgery). 9. Those who cannot guarantee sufficient time to participate in this study.
