研究目的: |
通过发掘与疾病分型相关的临床特征及多组学指标,构建基于激素疗效及长期预后疾病分型的预测模型,并通过多中心回顾性队列及前瞻性队列进行验证及优化,从而助力EGE患者的精准评估,为后续精准治疗打下坚实的基础。为了达成以上研究目的,课题组拟开展分布完成以下研究:首先,在前期发掘的EGE预后的临床特征及辅助检查指标的基础上,利用PUMCH-EGE队列构建疾病分型的临床预测模型;其次,利用已建成PUMCH-EGE的生物样本库开展多组学分析,构建疾病分型的多模态预测模型;再次,利用多中心的回顾性队列对前述的临床预测模型进行验证及优化;最后,利用多中心的前瞻性队列对优化的临床预测模型、多模态预测模型进行验证,从而建立并验证一套稳定、可靠的基于激素疗效及长期预后的EGE疾病分型预测体系,为实现该疾病的个体化治疗和精准医疗提供坚实的理论依据与实用的临床工具 |

Objectives of Study: |
This research aims to construct a predictive model for disease subtyping based on corticosteroid response and long-term prognosis by identifying clinical features and multi-omics biomarkers associated with disease subtypes. The model will be validated and optimized using multicenter retrospective and prospective cohorts, ultimately facilitating precise assessment of patients with eosinophilic gastroenteritis (EGE) and laying a solid foundation for subsequent precision treatment. To achieve these objectives, the research team plans to carry out the following studies step by step: First, based on previously identified clinical features and laboratory indicators associated with EGE prognosis, a clinical predictive model for disease subtyping will be established using the PUMCH-EGE cohort. Second, multi-omics analysis will be performed using the established PUMCH-EGE biobank to construct a multimodal predictive model for disease subtyping. Third, the aforementioned clinical predictive model will be validated and optimized using a multicenter retrospective cohort. Finally, the optimized clinical predictive model and the multimodal predictive model will be validated using a multicenter prospective cohort. Through these efforts, a stable and reliable predictive system for EGE disease subtyping based on corticosteroid response and long-term prognosis will be established and validated, providing a robust theoretical basis and practical clinical tool for individualized treatment and precision medicine for this disease. |

Exclusion criteria: |
1. Under 14 years old; 2. Suffering from parasitic infections, inflammatory bowel disease, eosinophilia, connective tissue diseases, malignant tumors, drug allergies and other diseases that cause eosinophilia; 3. Unwilling to participate in the study or unable to guarantee follow-up within 6 months after treatment. |
