研究目的:

目前,移植是末期疾病患者的终极治疗方法。基于免疫抑制方案在临床上广泛应用,并在移植患者中取得了显著成功,但部分患者仍存在移植物排斥和严重药物副作用。因此,治疗性药物监测(TDM)对于个体化和优化移植药物治疗至关重要。然而,仅基于 TDM 实现免疫抑制管理的精准性挑战巨大,因为移植药物血浆浓度的变化仅反映其体内药代动力学,无法直接反映移植患者的免疫抑制状态。因此急需一种单一简便的筛查方案如高通量的蛋白质组学和代谢组学,全面理解移植恢复过程中免疫状态动态以及不同临床结局患者之间的关系,有助于制定精确的临床管理策略(例如,在最佳时机为合适的患者预防移植物排斥和/或降低药物毒性)。  

Objectives of Study:

At present, transplantation is the ultimate treatment method for patients with end-stage diseases. The immunosuppressive regimen has been widely used in clinical practice and has achieved significant success in transplant patients, but some patients still suffer from transplant rejection and severe drug side effects. Therefore, therapeutic drug monitoring (TDM) is crucial for individualizing and optimizing transplant drug therapy. However, the precision challenge of implementing immunosuppressive management solely based on TDM is enormous, as changes in plasma concentration of transplant drugs only reflect their pharmacokinetics in vivo and cannot directly reflect the immunosuppressive status of transplant patients. Therefore, there is an urgent need for a single and simple screening protocol, such as high-throughput proteomics and metabolomics, to comprehensively understand the dynamics of immune status during transplant recovery and the relationship between patients with different clinical outcomes, which can help develop precise clinical management strategies (such as preventing transplant rejection and/or reducing drug toxicity for appropriate patients at the best time).

纳入标准:

Inclusion criteria

排除标准:

1、妊娠妇女或哺乳期妇女; 2、病情危急、难以对治疗的有效性和安全性做出确切评价者; 3、非肾脏移植手术; 4、其他联合移植手术。

Exclusion criteria:

1. Pregnant or lactating women; 2. Those with critical medical conditions and difficulty in accurately evaluating the effectiveness and safety of treatment; 3. Non renal transplant surgery; 4. Other combined transplantation surgeries.
