研究目的:

由于光遗传激活ZI GABA能神经元会使小鼠出现暴食性的进食行为。长时间激活ZI GABA能神经元促进小鼠进食增加,体重逐渐上升,停止光刺激后,小鼠体重就会下降回原水平。与此同时,我们发现光遗传激活自ZI GABA神经元投射至下游脑区VTA的神经末端,也能使小鼠进食量增加。这个结果与神经性贪食的病征十分类似,表现为进食动机增强诱发暴食行为导致肥胖。磁共振成像(MRI)技术,已被广泛应用于研究神经性贪食的大脑功能和结构变化。在神经性贪食患者中, ZI和VTA脑区是否参与病理性的发展过程是需要进一步探究的问题。在神经性贪食患者接受食物和非食物视觉刺激时,使用fMRI技术,可以准确的验证ZI和VTA 脑区对不同刺激的响应水平,检测ZI和VTA脑区的响应和其他脑区相比是否存在异质性,辨别ZI和VTA对食物视觉性刺激是否存在更高的选择性。  

Objectives of Study:

Optogenetic activation of ZI GABAergic neurons induces binge-like feeding behavior in mice. Prolonged activation of ZI GABAergic neurons promotes increased food intake and gradual body weight gain, while cessation of photostimulation causes body weight to return to baseline levels. At the same time, we found that optogenetic activation of the axon terminals of ZI GABAergic neurons projecting to the downstream brain region VTA also increases food intake in mice. These findings are highly similar to the clinical features of bulimia nervosa, in which enhanced feeding motivation triggers binge eating behavior and leads to obesity.Magnetic resonance imaging (MRI) has been widely used to investigate functional and structural brain alterations in bulimia nervosa. Whether the ZI and VTA are involved in the pathological progression of bulimia nervosa remains a question that requires further investigation. By using fMRI in patients with bulimia nervosa during exposure to food and non-food visual stimuli, it is possible to accurately assess the response levels of the ZI and VTA to different stimuli, examine whether the responses of the ZI and VTA differ from those of other brain regions, and determine whether the ZI and VTA show greater selectivity for food-related visual stimuli.

Inclusion criteria

排除标准:

1.精神活性物质或酒精等物质滥用者; 2.严重躯体残疾,无法完成随访者; 3.曾被诊断或共患其他符合ICD-10 或DSM-5 诊断标准的重性精神疾病、精神发育迟滞、痴呆及严重认知功能障碍者; 4.既往或目前患有任何影响脑部结构或功能的重大躯体疾病、神经系统疾病、脑外伤等; 5.有自杀倾向或不合作者; 6.身体内有金属植入,或其他不适用于磁共振扫描的情况; 7.妊娠或哺乳期女性; 8.颅内有金属、电子仪器、癫痫病史及家族史等TMS 禁忌症者; 9.其他研究者认为不适宜入组的检查异常。

Exclusion criteria:

1. Individuals with current substance abuse or dependence, including alcohol or other psychoactive substances; 2. Individuals with severe somatic illnesses that prevent completion of follow-up; 3. Individuals previously diagnosed with or co-occurring with other severe psychiatric disorders meeting ICD-10 or DSM-5 diagnostic criteria, intellectual disability, dementia, and severe cognitive impairment; 4. Individuals with current or past major somatic diseases, neurological disorders, or brain injuries that may affect brain structure or function; 5. Individuals with suicidal tendencies or poor cooperation; 6. Individuals with metallic implants in the body or other conditions that are not suitable for magnetic resonance imaging (MRI); 7. Pregnant or breastfeeding women; 8. Individuals with intracranial metal, electronic devices, a history of epilepsy, or a family history of epilepsy, or other contraindications for transcranial magnetic stimulation (TMS); 9. Other abnormal findings identified by the investigators that are considered unsuitable for inclusion in the study.

This study does not employ a dedicated electronic data capture (EDC) system. Study data will be collected using structured data recording forms and managed in password-protected electronic files stored on research team–designated computers or institutional servers. Data will be coded without direct personal identifiers, and access will be restricted to authorized study personnel only. Regular data backup will be performed to ensure data security and integrity.
