研究目的:

本项目旨在通过建立并分析子痫前期孕产妇临床队列,采集并检测其孕晚期粪便与血清样本,以达成以下目的: 鉴定特征性菌群与代谢物谱:明确子痫前期孕妇肠道菌群组成及血清代谢物的特异性改变。 建立关联网络:阐明上述菌群与代谢物异常与母体血清炎症因子(如TNF-α)水平升高的相关性。 筛选潜在生物标志物:构建能够区分子痫前期与正常妊娠的菌群-代谢物组合模型,为早期识别与机制研究提供依据。  

Objectives of Study:

This project aims to establish and analyze a clinical cohort of preeclamptic and normal pregnant women to achieve the following objectives through the collection and examination of their late-pregnancy fecal and serum samples:To identify characteristic microbiome and metabolite profiles: Determine the specific alterations in gut microbiota composition and serum metabolites in women with preeclampsia.To establish correlation networks: Elucidate the associations between these microbial/metabolite abnormalities and elevated levels of maternal serum inflammatory factors (e.g., TNF-α).To screen for potential biomarkers: Construct a combined microbiota-metabolite model capable of distinguishing preeclampsia from normal pregnancy, providing a basis for early identification and mechanistic research.

Inclusion criteria

排除标准:

1.多胎妊娠; 2.孕前存在慢性高血压、糖尿病、肾脏疾病; 3.近期使用抗生素或益生菌(4周内); 4.存在其他妊娠并发症; 5.拒绝签署知情同意书;

Exclusion criteria:

1.Multiple pregnancy;
2.Pre-existing chronic hypertension, diabetes, or kidney disease;
3.Recent use of antibiotics or probiotics (within 4 weeks);
4.Presence of other pregnancy complications;
5.Refusal to sign informed consent;

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

病例记录表:采用为本次回顾性队列研究专门设计的纸质版/电子版病例报告表,采集内容包括:研究对象的社会人口学信息、孕产史、子痫前期诊断标准相关的临床参数(血压、尿蛋白等)、样本采集时间及基本信息。 电子采集和管理系统:使用 REDCap 系统进行数据双录入、存储和管理。系统设置在安全的内网服务器,通过权限管理控制数据访问,确保数据的准确性、完整性和保密性。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture:

Case Record Form: A paper-based/electronic CRF specifically designed for this retrospective cohort study will be used to collect data, including: socio-demographic information, obstetric history, clinical parameters related to preeclampsia diagnosis (blood pressure, urinary protein, etc.), and sample collection time/basic information. Electronic Data Capture and Management: Data will be entered, stored, and managed using the REDCap system hosted on a secure internal server. Data accuracy, integrity, and confidentiality are ensured through double data entry and controlled access via user permissions.
