研究目的:

本课题拟取大肠癌患者的粪便样本、非肠癌组织样本及肠癌组织样本进行细菌测序,筛选并验证肿瘤归巢菌(大肠杆菌)的存在;随后分离肠癌组织和正常组织的大肠杆菌,通过动物实验和光学成像技术验证大肠杆菌的肿瘤靶向性;最后探索靶向肿瘤部位的大肠杆菌亚型的功能机制。本项目创新性地从肠道微生物的角度探索靶向大肠癌的肿瘤归巢菌,并解析其结构和功能。本项目为阐明大肠杆菌归巢特性提供了新的研究思路,为大肠癌的细菌疗法提供了新的工具菌株,具有重要的科学价值和临床意义。  

Objectives of Study:

This research project intends to collect fecal samples from colorectal cancer patients, non-cancerous tissue samples, and cancerous tissue samples for bacterial sequencing to screen and verify the presence of tumor-homing bacteria (Escherichia coli). Subsequently, Escherichia coli from colorectal cancer tissues and normal tissues will be isolated, and the tumor-targeting ability of Escherichia coli will be verified through animal experiments and optical imaging technology. Finally, the functional mechanism of the Escherichia coli subtype that targets tumor sites will be explored. This project innovatively explores tumor-homing bacteria targeting colorectal cancer from the perspective of intestinal microorganisms and analyzes their structure and function. It provides a new research idea for clarifying the homing characteristics of Escherichia coli and a new tool strain for bacterial therapy of colorectal cancer, thus having important scientific value and clinical significance.

Inclusion criteria

排除标准:

1.合并其他恶性肿瘤;
2.严重心肺疾病;
3.近3个月使用抗生素、激素或肠道菌群制剂;
4.合并克罗恩病、溃疡性结肠炎等其他肠道疾病;
5.精神疾病或认知、沟通功能障碍;

Exclusion criteria:

1.Complicated with other malignant tumors;
2.Severe cardiopulmonary diseases;
3.Use of antibiotics, hormones, or intestinal flora preparations in the past 3 months;
4.Combined with other intestinal diseases such as Crohn's disease and ulcerative colitis;
5.Mental illness or cognitive and communication dysfunction;
