研究目的：

本研究旨在建立SLE母子前瞻性家庭队列，通过系统收集并整合SLE母亲的人口学信息、疾病特征、围产期暴露因素、环境及行为暴露因素以及子代的长期随访数据，构建多维度生物信息数据库。本研究将ANA滴度≥1: 80定义为SLE患者子代发展为临床SLE的关键替代终点与血清学标志物，以精准识别处于“临床前自身免疫”阶段的SLE高风险个体，系统分析影响子代发病风险的关键因素。在此基础上，本研究利用机器学习算法，开发并验证一个具有临床转化意义的子代发病风险预测模型，旨在为育龄期女性SLE患者提供个体化的生育决策指导和科学的风险分层管理依据，填补该领域的研究空白。  

Objectives of Study：

This study aims to establish a prospective family cohort of SLE mothers and their children. By systematically collecting and integrating the demographic information, disease characteristics, perinatal exposure factors, environmental and behavioral exposure factors of SLE mothers, as well as the long-term follow-up data of their offspring, a multi-dimensional bioinformatics database will be constructed. In this study, an ANA titer of >= 1:80 was defined as a key surrogate endpoint and serological marker for the offspring of SLE patients to develop into clinical SLE, in order to accurately identify high-risk individuals of SLE in the "preclinical autoimmune" stage and systematically analyze the key factors influencing the risk of the offspring developing the disease. Based on this, this study utilized machine learning algorithms to develop and validate a offspring disease risk prediction model with clinical translational significance, aiming to provide individualized fertility decision-making guidance and scientific risk stratification management basis for female SLE patients of childbearing age, filling the research gap in this field.
