研究目的:

精神分裂症(Schizophrenia, SCH)是一种严重的精神障碍,全球患病率约1%。截止2022年,全球精神分裂症患者已超过2400万人。尽管随着抗精神病药物的不断问世,患者的临床症状得到了较好的控制,但过早衰老现象在精神分裂症患者中仍普遍存在且形势严峻。精神分裂症患者的早衰现象不仅缩短了患者的预期寿命,且导致了更高的全因死亡率,研究发现,精神分裂症患者全因死亡率是普通人群的2-2.5倍,主要与年龄相关疾病的高发生率有关,且心血管疾病造成的寿命损失最大。过早的衰老使得精神分裂症患者在生活质量和生理健康方面面临严重挑战,同时给其家庭和社会带来了沉重的负担。因此,本课题通过比较分析精神分裂症患者和健康对照人群的DNA甲基化、端粒长度、衰老相关分泌表型水平,同时建立精神分裂症患者长程随访队列,分析精神分裂症患者DNA甲基化、端粒长度、衰老相关分泌表型水平的纵向变化,深入系统研究精神分裂症患者过早衰老的现状,寻找导致精神分裂症过早衰老的危险因素并探索与其过早衰老相关的生物学机制,为改善患者预后和生活质量以及制定更有效的干预措施提供科学依据。  

Objectives of Study:

Schizophrenia is a severe mental disorder with a global prevalence of approximately 1%. By 2022, the number of schizophrenia patients worldwide had exceeded 24 million. Although the continuous development of antipsychotic medications has led to better control of clinical symptoms, premature aging remains a widespread and serious issue among schizophrenia patients. This phenomenon not only shortens the life expectancy of patients but also leads to a higher all-cause mortality rate. Studies have found that the all-cause mortality rate among schizophrenia patients is 2 to 2.5 times higher than that of the general population, primarily due to the increased incidence of age-related diseases, with cardiovascular diseases contributing the most to reduced lifespan. Premature aging poses significant challenges to the quality of life and physical health of schizophrenia patients, while also placing a heavy burden on their families and society. Therefore, this study aims to systematically investigate premature aging in schizophrenia patients by comparing and analyzing DNA methylation, telomere length, and senescence-associated secretory phenotype (SASP) levels between schizophrenia patients and healthy controls. Additionally, a long-term follow-up cohort of schizophrenia patients will be established to examine longitudinal changes in these biomarkers. The research seeks to identify risk factors contributing to premature aging in schizophrenia and explore the underlying biological mechanisms, thereby providing a scientific basis for improving patient prognosis and quality of life, as well as for developing more effective intervention strategies.

1.共患其他精神疾病; 2.妊娠或哺乳期患者; 3.罹患终末期疾病,包括肾衰、心衰、癌症等。

1.Comorbid presence of other psychiatric disorders; 2.Pregnancy or lactation; 3.Diagnosis of terminal illnesses, including renal failure, heart failure, cancer, etc.
