1.主要目的: 探索颅骨骨髓介导的神经免疫轴在药物难治性癫痫发生发展中的作用机制,识别与癫痫发作频率、严重程度或药物难治性癫痫相关的关键免疫细胞亚群和分子通路。 2.次要目的: (1) 比较药物难治性癫痫患者与对照组颅骨骨髓、脑膜、脉络丛和癫痫灶的免疫细胞组成和基因表达谱差异。 (2) 分析颅骨骨髓、脑膜、脉络丛和癫痫灶之间免疫细胞亚群组成与迁移相关分子表达谱的关联性,探索颅骨骨髓作为免疫细胞潜在来源的证据及其与癫痫病理的潜在联系。 (3) 评估特定免疫细胞亚群或细胞因子在药物难治性癫痫诊断、预后评估及治疗靶点开发中的潜力。 (4) 为开发基于神经免疫轴的药物难治性癫痫新型干预策略(如靶向免疫治疗或细胞治疗)提供理论依据。 |

Objectives of Study: |
1. Primary Objective: To investigate the role of the skull bone marrow-mediated neuro-immune axis in the pathogenesis of drug-resistant epilepsy, and to identify key immune cell subsets and molecular pathways associated with seizure frequency, disease severity, or drug resistance. 2. Secondary Objectives: (1) To compare the composition and transcriptomic profiles of immune cells in skull bone marrow, meninges, choroid plexus, and epileptogenic foci between patients with drug-resistant epilepsy and non-epilepsy controls. (2) To analyze the associations between immune cell populations and migration-related molecular signatures across skull bone marrow, meninges, choroid plexus, and epileptic foci, to evaluate skull bone marrow as a potential source of pathogenic immune cells. (3) To assess the diagnostic, prognostic, and therapeutic potential of specific immune cell subsets or cytokines in drug-resistant epilepsy. (4) To provide a mechanistic foundation for developing novel neuro-immune-targeted interventions (e.g., immunomodulatory or cell-based therapies) for drug-resistant epilepsy. |

1. 病例组(药物难治性癫痫组)排除标准: (1) 合并其他严重神经系统疾病(如脑肿瘤、脑卒中、多发性硬化等)。 (2) 合并严重免疫系统疾病或自身免疫病。 (3) 患有严重全身性感染或炎症性疾病。 (4) 正在接受可能影响免疫反应的治疗(如化疗、放疗、免疫抑制剂治疗等) (5) 妊娠或哺乳期妇女。 (6) 有精神疾病史,无法配合研究。 2. 对照组(非癫痫组)排除标准: (1) 合并严重免疫系统疾病或自身免疫病。 (2) 患有严重全身性感染或炎症性疾病。 (3) 正在接受可能影响免疫反应的治疗。 (4) 妊娠或哺乳期妇女。 (5) 有精神疾病史,无法配合研究。 |

1. Exclusion criteria for the case group (drug-resistant epilepsy group): (1) Coexisting other severe neurological diseases (such as brain tumor, stroke, multiple sclerosis, etc.). (2) Coexisting severe immune system diseases or autoimmune disorders. (3) Suffering from severe systemic infection or inflammatory disease. (4) Receiving treatments that may affect immune response (such as chemotherapy, radiotherapy, immunosuppressive therapy, etc.). (5) Pregnant or lactating women. (6) History of psychiatric illness, unable to cooperate with the study. 2. Exclusion criteria for the control group (non-epilepsy): (1) Coexisting severe immune system diseases or autoimmune disorders. (2) Suffering from severe systemic infection or inflammatory disease. (3) Receiving treatments that may affect immune response. (4) Pregnant or lactating women. (5) History of psychiatric illness, unable to cooperate with the study. |
