研究目的：

基于单细胞转录组测序构建胃癌前病变动态演变图谱，找到三型肠化生发展为胃癌的关键机制和特征分子，并通过人胃类器官模型进行验证，为胃癌的早期诊断和治疗提供新的分子依据，帮助医生及早发现高危人群，从而降低胃癌的发生风险。  

Objectives of Study：

We are using single-cell transcriptome sequencing to construct a dynamic evolutionary map of gastric precancerous lesions. In doing so, we identify the key mechanisms and characteristic molecules underlying the progression of the three types of intestinal metaplasia to gastric cancer. These findings are then validated using human gastric organoid models. Our work provides new molecular evidence for the early diagnosis and treatment of gastric cancer, helping clinicians identify high-risk populations earlier and thereby reducing the incidence of gastric cancer.

纳入标准：

Inclusion criteria

排除标准：

1. 合并其他恶性肿瘤，伴有严重的心、肝、肾等主要脏器病变，或严重 的血液、神经、精神类疾病； 2. 半年内做过幽门螺杆菌根除治疗； 3. 既往胃部放疗/化疗史。

Exclusion criteria：

1. Concomitant other malignant tumors, severe dysfunction of major organs (heart, liver, kidney, etc.), or serious hematological, neurological, or psychiatric diseases. 2. Helicobacter pylori eradication therapy within the past six months. 3. Prior gastric radiotherapy or chemotherapy.

电子病历系统采集，数据上传至GEO公共数据库启用“受控访问”模式（需审批后获取），采用三级编码系统（样本编码、临床信息编码、基因数据编码独立存储）防止临床信息关联暴露。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：

Data are extracted from the electronic medical-record system and uploaded to the GEO public database under "Controlled-Access" mode (available only after approval). A three-level coding system—specimen ID, clinical-info ID, and genomic-data ID—is stored separately to prevent linkage and re-identification of clinical information.
