研究目的:

分析膜性肾病与健康对照者肾脏中MT-ND6、MT-ND6下游以及炎症信号通路相关分子表达情况以及MT-ND6 mRNA的m6A甲基化水平,探究MT-ND6以及MT-ND6 mRNA的m6A甲基化水平在膜性肾病发病机制中的作用。  

Objectives of Study:

Analyze the expression of MT-ND6, downstream MT-ND6, and inflammatory signaling pathway related molecules in the kidneys of membranous nephropathy and healthy controls, as well as the m6A methylation level of MT-ND6 mRNA, to explore the role of MT-ND6 and m6A methylation level of MT-ND6 mRNA in the pathogenesis of membranous nephropathy.

纳入标准:

Inclusion criteria

排除标准:

膜性肾病组排除标准: 1.继发性MN(如SLE、HBV感染、恶性肿瘤相关); 2.合并其他原发性肾小球疾病(如IgA肾病); 3.肾活检肾小球数<10个或病理资料不全; 4.妊娠期或哺乳期女性; 5.合并其他可能危及生命的疾病; 正常对照组排除标准: 1.不具备肾脏手术治疗指征。

Exclusion criteria:

Exclusion criteria for membranous nephropathy group: 1. Secondary MN (such as SLE, HBV infection, related to malignant tumors); 2. Coexisting other primary glomerular diseases (such as IgA nephropathy); 3. Renal biopsy showing less than 10 glomeruli or incomplete pathological data; 4. Pregnant or breastfeeding women; 5. Coexisting other potentially life-threatening diseases; Exclusion criteria for normal control group: 1. No indications for surgical treatment of the kidney.

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

收集患者行肾穿刺活检/手术治疗前3天内的相关数据为基线数据,由两人独立收集并记录在CRF表格中,随后由数据管理员检查条目的一致性,对于不一致的条目,由三人共同核查,随后录入数据库, 对于每次录入的数据随机20%进行再次核查。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture:

Collect relevant data from patients within 3 days before undergoing renal biopsy/surgical treatment as baseline data, independently collected by two individuals and recorded in the CRF form. Then, the data administrator checks the consistency of the entries. For inconsistent entries, three individuals jointly verify and then enter them into the database. For each entry, 20% of the data is randomly rechecked.
