研究目的：

本研究开发了一种利用脐血为来源生产CD73+NK细胞的方法，在短时间内便能制备出具有定向作用的CD73+NK细胞，增强NK细胞的特异性功能，能给临床治疗提供一定的技术支持。 1.本研究提供了一种脐血造血干细胞分离工艺，采用6%羟乙基淀粉密度梯度离心法是相对理想的脐带血造血干细胞分离方法，并利用响应面法对离心温度，离心力，离心时间三个影响因素进行了分析优化，分离的HSCs细胞活力可达到最佳状态; 2.本研究研制了一种针对不同起始来源的脐血造血干细胞诱导培养的CD73+NK方案，为临床治疗用细胞提供了新参考制备方案; 3.本研究初步阐明CD73+NK特异性功能，为NK细胞的亚型筛选提供新依据，推动NK细胞治疗效果评价的深入。  

Objectives of Study：

In this study, a method for producing CD73 NK cells using umbilical cord blood as a source can prepare CD73 NK cells with directional effects in a short period of time, enhance the specific function of NK cells, and provide certain technical support for clinical treatment. 1. This study provides a process for isolation of cord blood hematopoietic stem cells, using 6% hydroxyethyl starch density gradient centrifugation method is a relatively ideal method for isolation of cord blood hematopoietic stem cells, and the response surface method is used to analyze and optimize the three influencing factors of centrifugation temperature, centrifugal force and centrifugation time, and the viability of the isolated HSCs cells can achieve the best state. 2. In this study, a CD73 NK regimen for umbilical cord blood hematopoietic stem cell induction culture was developed for different starting sources, which provided a new reference preparation scheme for cells for clinical treatment. 3. This study preliminarily elucidated the specific function of CD73 NK, provided a new basis for NK cell subtype screening, and promoted the in-depth evaluation of NK cell therapy effect.

Inclusion criteria

排除标准：

1.新生儿父母双方有遗传病史(三代以内有家族血液病史)、有传染病史、有吸毒史、新生儿父母双方有冶游史、12 个月内有输血史及血液制品使用史； 2.怀孕期间各项检查指标明显异常，包括但不限于产妇血红蛋白＜90g/L，收缩压>=140 mmHg（1 mmHg=0.133 kPa）和/或舒张压>=90 mmHg，OGTT 服糖后 2 小时血糖 >= 11.1 mmol/L 等；出现孕期并发症，包括但不限于妊娠期糖尿病、妊娠期高血压综合症、妊娠期肝内胆汁淤积综合症、胎盘早剥、产科弥漫性血管内凝血等； 3.8 周内曾接受减毒活疫苗注射，如伤寒疫苗、风疹活疫苗、狂犬病疫苗、水痘疫苗。

Exclusion criteria：

1. Both parents of the newborn have a history of genetic diseases (family blood history within three generations), a history of infectious diseases, a history of drug abuse, a history of metallurgy for both parents of the newborn, a history of blood transfusion and the use of blood products within 12 months; 2. Obvious abnormalities in various examination indicators during pregnancy, including but not limited to maternal hemoglobin <90g/L, systolic blood pressure >=140 mmHg (1 mmHg=0.133 kPa) and/or diastolic blood pressure >=90 mmHg, blood glucose >= 11.1 mmol/L at 2 hours after taking sugar in OGTT, etc.; Pregnancy complications, including but not limited to gestational diabetes, gestational hypertension syndrome, gestational intrahepatic cholestasis syndrome, placental abruption, obstetric diffuse intravascular coagulation, etc.; 3. Have received live attenuated vaccine injections within 8 weeks, such as typhoid vaccine, live rubella vaccine, rabies vaccine, and chickenpox vaccine.
