研究目的：

建立肝癌、胆管癌和胰腺癌三个癌种的类器官标准化培养技术体系，及类器官的组织学、遗传学表征实验体系；进行肿瘤类器官用于临床个体化治疗的初步研究，为注册申请临床试验获取必要的数据支持；基于肿瘤组织和配对癌旁组织/完全正常组织的基因组、转录组和蛋白组等多组学检测及分析，预期发现潜在的新型诊疗靶点及策略；对病人的血浆中外泌体和相应肿瘤类器官的外泌体进行提取和代谢物分析，预期发现潜在的早期诊断标志物及策略。  

Objectives of Study：

To establish standardized organoid culture systems for three major cancer types: hepatocellular carcinoma, cholangiocarcinoma, and pancreatic cancer, along with organoid-based histological and genetic characterization platforms. Preliminary studies will be conducted to explore the clinical application of tumor organoids in personalized treatment, aiming to generate essential data for future clinical trial registration. By performing multi-omics analyses—including genomics, transcriptomics, and proteomics—on tumor tissues and matched adjacent non-tumorous or normal tissues, the study aims to identify novel diagnostic and therapeutic targets and strategies. In addition, exosomes from patient plasma and corresponding tumor organoids will be isolated and subjected to metabolomic profiling to discover potential biomarkers and strategies for early cancer detection.
