基于精神分裂症家系研究，量化环境暴露（例如童年逆境、贫穷、父亲高龄和物质使用等）对疾病代际传递的影响，系统分析环境暴露、临床特征与多组学特征的关联性，探索精神分裂症的内表型，识别与疾病转归关联的环境-遗传-内表型路径.

Objectives of Study：

Based on family-based studies of schizophrenia, this research aims to quantify the contributions of environmental exposures (e.g., childhood adversity, poverty, advanced paternal age, and substance use) to the intergenerational transmission of the disorder; systematically analyze the associations among environmental exposures, clinical charac -teristics, and multi-omics profiles; explore endophenotypes of schizophrenia; and identify environment-genetics-endophenotype pathways associated with disease outcomes.

Inclusion criteria

1. Schizophrenia Families with Intergenerational Transmission:Offspring Requirements:
(1) Aged between 16 and 45 years (inclusive);(2) Has one biological parent diagnosed with schizophrenia;(3) Diagnosed with schizophrenia following assessment using the Structured Clinical Interview for DSM-IV-TR Axis I Disorders, Research Version (SCID-I/P);(4) Capable of comprehending interview content and providing written informed consent.Parent Requirements: One parent diagnosed with schizophrenia, and the non-affected parent capable of comprehending interview content and providing written informed consent.
2. Schizophrenia Families without Intergenerational Transmission:Offspring Requirements:(1) Aged between 16 and 45 years (inclusive);(2) Has one biological parent diagnosed with schizophrenia;(3) Receives no psychiatric diagnosis following assessment using the Structured Clinical Interview for DSM-IV-TR Axis I Disorders, Research Version (SCID-I/P);(4) Capable of comprehending interview content and providing written informed consent. Parent Requirements: One parent diagnosed with schizophrenia, and the non-affected parent capable of comprehending interview content and providing written informed consent.
3. Healthy Control Group without Family Psychiatric History:
Requirements:(1) Aged between 16 and 45 years (inclusive);(2) Negative family history of psychiatric disorders;(3) Receives no psychiatric diagnosis following assessment using the Structured Clinical Interview for DSM-IV-TR Axis I Disorders, Research Version (SCID-I/P);(4) Capable of comprehending interview content and providing written informed consent.

排除标准：

1.精神分裂症疾病代际传递家系：（1）精神分裂症患者子女一方亲代明确诊断精神分裂症外，另一方家长曾有以下任一精神疾病史：精神分裂症、分裂情感障碍、妄想性障碍、精神发育迟滞、双相情感障碍、重度抑郁发作或癫痫所致的精神障碍；（2）研究对象存在脑器质性疾病及癫痫发作史；（3）研究对象既往和现患严重躯体疾病；（4）研究对象存在严重认知障碍等研究者判断不适宜参与研究的情况。 2.精神分裂症疾病未发生代际传递家系：（1）精神分裂症患者子女一方亲代明确诊断精神分裂症外，另一方家长曾有以下任一精神疾病史：精神分裂症、分裂情感障碍、妄想性障碍、精神发育迟滞、双相情感障碍、重度抑郁发作或癫痫所致的精神障碍；（2）研究对象存在脑器质性疾病及癫痫发作史；（3）研究对象既往和现患严重躯体疾病；（4）研究对象存在严重认知障碍等研究者判断不适宜参与研究的情况。 3:健康对照（1）存在高血压、糖尿病、高血脂、自身免疫性疾病等慢性系统性疾病；（2）存在酒精或药物滥用/依赖（如大麻、可卡因、苯丙胺等）；（3）精神科药物（如抗抑郁药、抗精神病药、镇静剂等）使用史；（4）左利手；（5）怀孕或哺乳期；（6）无法配合完成研究流程（如语言障碍、文化程度不足）。

Exclusion criteria：

1. Schizophrenia Families with Intergenerational Transmission:(1) Aside from the biological parent definitively diagnosed with schizophrenia, the other parent has a history of any of the following psychiatric disorders: schizophrenia, schizoaffective disorder, delusional disorder, intellectual disability (mental retardation), bipolar disorder, major depressive episode, or mental disorders due to epilepsy;(2) Participants have a history of organic brain diseases or epilepsy/seizure disorders;(3) Participants have a history of or currently suffer from severe physical illnesses;(4) Participants have severe cognitive impairment or other conditions deemed by investigators to render participation unsuitable. 2. Schizophrenia Families without Intergenerational Transmission:(1) Aside from the biological parent definitively diagnosed with schizophrenia, the other parent has a history of any of the following psychiatric disorders: schizophrenia, schizoaffective disorder, delusional disorder, intellectual disability (mental retardation), bipolar disorder, major depressive episode, or mental disorders due to epilepsy;(2) Participants have a history of organic brain diseases or epilepsy/seizure disorders;(3) Participants have a history of or currently suffer from severe physical illnesses;(4) Participants have severe cognitive impairment or other conditions deemed by investigators to render participation unsuitable. 3. Healthy Control Group:(1) Presence of chronic systemic diseases (e.g., hypertension, diabetes, hyperlipidemia, autoimmune diseases);(2) History of alcohol or substance abuse/dependence (e.g., cannabis, cocaine, amphetamines);(3) History of psychotropic medication use (e.g., antidepressants, antipsychotics, sedatives);(4) Left-handedness; (5) Pregnancy or lactation;(6) Inability to cooperate with or complete study procedures (e.g., due to language barriers, insufficient education/cultural factors).

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：	研究中采用电子数据的方式进行采集。临床访谈及执行功能评估等纸质版资料将采用Epidata 录入和管理，在原始资料收集一周内录入电子数据库，数据不完整或有疑问需及时补充核实。建立受试者资料档案进行临床档案管理，所有档案按编号、类别分装入资料盒后，保存在有锁的档案柜中，仅供研究人员查阅。
Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：	Electronic data capture (EDC) methods were employed for data collection throughout the study. Paper-based records, including clinical interviews and executive function assessments, were entered and managed using Epidata software. Data entry into the electronic database was completed within one week of collecting the original records. Any incomplete or questionable data points were promptly supplemented and verified. Subject records were compiled into individual clinical files for management. All files were systematically organized by participant ID and category, stored in designated archive boxes, and secured within locked (and preferably fireproof) filing cabinets. Access to these files was restricted to authorized research personnel.
