研究目的：

1. 评价GYS-016作为结核病宿主导向治疗（HDT）药物，辅助治疗耐药肺结核的安全性和初步疗效，并探索GYS-016的最佳用药疗程； 2. 探究GYS-016辅助治疗耐药结核病对宿主免疫系统的影响及其机制，为结核病的HDT治疗提供创新思路； 3. 建立一种以结核病抗菌治疗为主、GYS-016 HDT为辅的全新综合治疗策略，最终应用于临床，真正做到从实验室到临床的转化。

Objectives of Study：

1. To evaluate the safety and preliminary efficacy of GYS-016 as a host-directed therapy (HDT) agent adjunctive to drug-resistant pulmonary tuberculosis treatment, and to explore the optimal treatment duration of GYS-016. 2. To investigate the effects and mechanisms of GYS-016 adjunctive therapy on the host immune system in drug-resistant tuberculosis, providing innovative insights for HDT in tuberculosis management. 3. To establish a novel comprehensive treatment strategy combining antitubercular therapy as the primary approach with GYS-016 HDT as an adjunct, ultimately achieving clinical translation from bench to bedside.

Inclusion criteria

排除标准：

1.既往对方案中使用的任何药物有过敏反应史； 2.合并严重的肝、肾、代谢、自身免疫性疾病、神经、精神或内分泌疾病、血液系统疾病、恶性肿瘤，以及长期服用免疫抑制剂药物者； 3.妊娠； 4.经治疗前评估不适宜参加本项目； 5.参加其它临床试验。

Exclusion criteria：

1. History of allergic reactions to any drugs used in the study protocol. 2. Concomitant severe medical conditions, including: Hepatic, renal, metabolic, or autoimmune disorders Neurological, psychiatric, or endocrine diseases Hematologic malignancies or active cancers Long-term use of immunosuppressive agents. 3. Pregnancy. 4. Deemed ineligible for participation after baseline evaluation. 5. Current enrollment in other clinical trials.
