研究目的:

1.临床疗效比较:通过系统监测探诊出血、探诊深度、附着丧失等关键临床指标,详细对比半导体激光辅助治疗和盐酸米诺环素药物辅助治疗慢性牙周炎的临床疗效。 2.微生物群落分析:聚焦不同辅助治疗方式在治疗前后不同时间点对口腔龈下菌斑微生物群落结构和多样性的影响,全面解析微生物群落的组成、丰度变化,探究优势菌群的演替规律,以及不同治疗方式对微生物群落的影响。 3.关联探究:深入探究临床疗效与口腔微生物群落变化之间的内在关联,试图揭示不同辅助治疗方式改善牙周状况的潜在微生物学机制,为慢性牙周炎的个性化治疗提供理论支持和实践指导。  

Objectives of Study:

1. Comparison of clinical efficacy: By systematically monitoring key clinical indicators such as bleeding on probing, probing depth, and attachment loss, the clinical efficacy of semiconductor laser - assisted treatment and minocycline hydrochloride drug - assisted treatment for chronic periodontitis will be compared in detail. 2. Microbiome analysis: Focusing on the impact of different assisted treatment methods on the structure and diversity of the subgingival plaque microbiome at different time points before and after treatment, comprehensively analyze the composition and abundance changes of the microbiome, explore the succession rules of dominant flora, and study the effects of different treatment methods on the microbiome. 3. Correlation exploration: Deeply explore the internal correlation between clinical efficacy and changes in the oral microbiome, attempt to reveal the potential microbiological mechanisms by which different assisted treatment methods improve periodontal conditions, and provide theoretical support and practical guidance for the personalized treatment of chronic periodontitis.

Inclusion criteria

排除标准:

1.全身情况差,无法耐受牙周基础治疗;存在牙周基础治疗、激光治疗和药物治疗的禁忌症。

Exclusion criteria:

1.Patients with poor general conditions are unable to tolerate periodontal basic treatment;
2.there are contraindications to periodontal basic treatment, laser treatment, and drug treatment.

In the research design, the random number sequence was generated by Li Xiaoqing, the principal investigator of this study, using R software. Specifically, in R software, relevant random functions such as the `runif()` function were used. 数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

数据采集通过设计问卷收集临床背景信息,由经统一培训的口腔科医生测量探诊出血、牙周袋深度和临床附着丧失程度,于治疗前后多个时间点采集数据,同时采集双侧龈下菌斑送专业机构对 V3 - V4 区开展 16S rRNA 基因测序。数据管理方面,设计录入表格,采用双人双录入比对修正 。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture:

Data collection is carried out by designing questionnaires to collect clinical background information. Uniformly trained stomatologists measure bleeding on probing, periodontal pocket depth, and clinical attachment loss. Data is collected at multiple time points before and after treatment. Meanwhile, bilateral subgingival plaque is collected and sent to a professional institution for 16S rRNA gene sequencing of the V3 - V4 region. In terms of data management, data entry forms are designed, and a double - entry method by two people is adopted for comparison and correction.
