研究目的: |
肺动脉高压(PAH)被称为心血管疾病的“恶性肿瘤”,进展快、死亡率高,严重危害国民健康,造成极大的社会和家庭负担。目前,PAH发病机制不清,肺小血管重构被认为是主要原因。PAH患者严重依赖药物治疗,缺乏阻断疾病进展的特效药物和策略。新近研究发现PAH骨髓具有类似骨髓增殖性肿瘤病理改变,且移植健康骨髓能够改善PAH的症状,提示骨髓源性细胞可能参与PAH发生。前期我们采用单细胞转录组学结合深度生信分析,在PAH模型中围绕骨髓、脾脏等多个组织开展了研究,初步绘制了PAH多器官单细胞图谱,发现了PAH骨髓和心肌组织中特有的MDSCs细胞亚群。因此,本课题拟基于上述发现,采用深度生信分析结合谱系追踪,深入探究PAH患者骨髓干细胞龛变化规律和转分化特点,筛选PAH中干细胞龛中效应细胞,揭示PAH发病分子机制,为PAH的防治提供新靶点和策略。 |

Objectives of Study: |
Pulmonary arterial hypertension (PAH) is known as the "malignant tumor" of cardiovascular diseases, with rapid progression and high mortality, which seriously endangers national health and causes great social and family burden. At present, the pathogenesis of PAH is unclear, and pulmonary small vessel remodeling is considered to be the main cause. Patients with PAH are heavily dependent on medication and lack specific drugs and strategies to block disease progression. Recent studies have found that PAH bone marrow has pathological changes similar to myeloproliferative tumors, and transplantation of healthy bone marrow can improve the symptoms of PAH, suggesting that bone marrow-derived cells may be involved in PAH occurrence. In the early stage, we used single-cell transcriptomics combined with deep biogenic analysis to carry out studies in multiple tissues such as bone marrow and spleen in PAH model, initially drew the multi-organ single-cell map of PAH, and found the specific MDSCs cell subsets in the bone marrow and cardiac tissue of PAH. Therefore, based on the above findings, this project intends to use deep biogenic analysis combined with lineage tracking to deeply explore the change rules and transdifferentiation characteristics of bone marrow stem cell niches in patients with PAH, screen effector cells in stem cell niches in PAH, reveal the molecular mechanism of pathogenesis of PAH, and provide new targets and strategies for prevention and treatment of PAH. |

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture: |
Data were collected and registered by the inpatient management system and surgical patient management system of the Second Affiliated Hospital of Army Military Medical University. Clinical cases were converted into case report forms, and paper versions of the report forms and experimental records were placed in the Laboratory of Cardiovascular Medicine of the Second Affiliated Hospital for safekeeping. Special personnel in the laboratory will archive and manage the electronic and paper versions of patient clinical data case report forms and experimental records. |
