研究目的:

小胶质细胞活化是帕金森病(PD)的核心病理标志。在促炎激活后,小胶质细胞将代谢从氧化磷酸化(OXPHOS)转向糖酵解。糖酵解驱动的乳酸积累有助于PD的发展和进展。然而,其潜在机制尚不清楚。这项工作共同证明了乳酸/Dld-K127/丙酮酸正反馈回路在“代谢-表观遗传”水平上驱动DA神经元损伤,并表明抑制这种恶性循环是治疗PD的有前景的治疗靶点。  

Objectives of Study:

Microglial activation is a central pathological hallmark of Parkinson's disease (PD). Microglia switch metabolism from oxidative phosphorylation (OXPHOS) toward glycolysis upon pro-inflammatory activation. Glycolysis-drived lactylate accumulation contributed to the development and progression of PD. However, the underlying mechanisms remain unclear. This work demonstrated a lactate/Dld-K127/pyruvate positive feedback loop that drived DA neuronal damage in "metabolism - epigenetic" level and suggested that suppressing this vicious feedback circle as a promising therapeutic target for PD

Inclusion criteria

排除标准:

HCs排除标准:(1)患有焦虑、抑郁等严重精神类疾病;(2)MMSE评分<24分。PD患者排除标准:(1)药物诱发的帕金森综合征、多系统萎缩、血管源性帕金森综合征等其他神经系统疾病;(2)有严重精神分裂等精神疾病;(3)合并颅内器质性病变或颅脑损伤;(4)合并心脏、肝脏、肾脏等器质性病变。

Exclusion criteria:

Exclusion criteria for HCs: (1) Suffering from severe mental illnesses such as anxiety and depression; (2) MMSE score<24 points. Exclusion criteria for PD patients:(1) Drug induced Parkinson's syndrome, multiple system atrophy, vascular Parkinson's syndrome, and other neurological disorders; (2) Having serious mental illnesses such as schizophrenia; (3) Combined intracranial organic lesions or traumatic brain injury; (4) Merge organic lesions such as heart, liver, and kidney.
