研究目的：

（1）探究二甲双胍与塞来昔布单独或联合使用对于FAP的临床具体疗效，评估二甲双胍与塞来昔布联合使用对于FAP患者抑制息肉生成是否具有协同作用。（2）探究二甲双胍是否通过影响TGF-β/PI3K/AKT信号转导来抑制FAP腺瘤性息肉的生长，以明确二甲双胍在抑制腺瘤性息肉生长方面的具体作用机制。（3）通过建立人工智能辅助系统来协助内镜下鉴别FAP进展期腺瘤及其癌变情况。

Objectives of Study：

(1) To investigate the clinic-specific efficacy of metformin and celecoxib alone or in combination for FAP, and to assess whether the combination of metformin and celecoxib has a synergistic effect on inhibiting polypogenesis in patients with FAP. (2) To investigate whether metformin inhibits the growth of adenomatous polyps in FAP by affecting TGF-β/PI3K/AKT signaling, in order to clarify the specific mechanism of metformin in inhibiting the growth of adenomatous polyps. (3) To assist endoscopic identification of FAP progressive adenomas and their carcinomas by establishing an artificial intelligence-assisted system.

纳入标准：

Inclusion criteria

排除标准：

I.对非甾体抗炎药、磺胺类药物以及水杨酸类药物过敏； II.C-反应蛋白升高(＞3.0 mg/L)； III.有心血管疾病史或出现以下任意一种情况，包括心肌梗塞、心绞痛、冠状动脉血管成形术、充血性心力衰竭、中风或冠状动脉搭桥手术、高血压未得有效到控制； IV.在试验前6个月内使用非甾体抗炎药每周 3 次以上、长期使用糖皮质激素； V.诊断为恶性疾病，如CRC； VI.诊断为糖尿病、怀孕或哺乳； VII.肝功能、肾功能检查结果显著异常，或有重大感染、呼吸道疾病、消化性溃疡等； VIII.备孕的男性； IX.研究者认为存在有不合适入选因素的患者。

Exclusion criteria：

I. Hypersensitivity to NSAIDs, sulfonamides, and salicylates; II.Elevated C-reactive protein (>3.0 mg/L); III. History of cardiovascular disease or any of the following conditions, including myocardial infarction, angina pectoris, coronary angioplasty, congestive heart failure, stroke or coronary artery bypass grafting surgery, and hypertension that is not effectively controlled; IV. Use of non-steroidal anti-inflammatory drugs more than 3 times per week, long-term use of glucocorticoids during the 6 months prior to the trial; V. Diagnosis of malignant disease, such as CRC; VI. Diagnosis of diabetes mellitus, pregnancy or breastfeeding; VII. Significantly abnormal results of liver and kidney function tests, or major infections, respiratory diseases, peptic ulcers, etc; VIII.Men who are preparing for pregnancy; IX. Patients who, in the opinion of the investigator, have inappropriate enrollment factors.
