研究目的：

MDD 与失眠常发生于同一患者，并伴随着大脑功能的改变，加重患者症状，降低治疗效果。而目前 MDD 伴失眠的病理机制研究尚不充分。因此，本研究通过静息态功能磁共振利用功能连接分析方法，进行一横断面研究，探索伴与不伴失眠的 MDD 患者的反刍思维和默认网络功能连接的差异。通过比较伴失眠的 MDD 患者与不伴失眠的 MDD 患者、健康人反刍思维和默认网络功能连接的差异，从而揭示MDD 伴失眠患者的神经生物学机制。为 MDD 伴失眠患者提供潜在的治疗靶点具有重要的临床意义和研究价值。

Objectives of Study：

MDD and insomnia often occur in the same patient and are accompanied by changes in brain function, which aggravate the patient's symptoms and reduce the effectiveness of treatment. At present, the pathological mechanisms of MDD with insomnia have not been adequately studied. Therefore, in this study, we conducted a cross-sectional study by resting-state functional magnetic resonance (MRI) using functional connectivity analysis to explore the differences in the functional connectivity of ruminative thinking and default network in MDD patients with and without insomnia. By comparing the differences in rumination and default network functional connectivity between MDD patients with insomnia, MDD patients without insomnia, and healthy individuals, the neurobiological mechanisms of MDD with insomnia will be revealed. It is of great clinical significance and research value to provide potential therapeutic targets for MDD patients with insomnia.

纳入标准：

Inclusion criteria

1) MDD with insomnia group:
① Age 18-65 years old, right-handed, male and female;
② Adequate comprehension ability, education level ≥ 9 years;
③ Meet the diagnostic criteria of DSM-5 Major Depressive Disorder, and SCID-5 confirms the diagnostic results;
④ PSQI score ≥ 7;
⑤ Understand the content of the study and sign the informed consent form.
(2) MDD without insomnia group:
① Age 18-65 years old, right-handed, men and women are not limited;
① Age 18-65 years old, right-handed, male or female; ② Sufficient understanding ability, education level ≥ 9 years;
③ Meet the diagnostic criteria of DSM-5 Major Depressive Disorder, and SCID-5 confirms the diagnosis;
④ PSQI score < 7;
⑤ Understand the content of the study and sign the informed consent form.
3) Healthy control group:
① Age 18-65 years old, right-handed, men and women are not limited;
② Adequate comprehension ability, education level ≥ 9 years;
(iii) SCID-5 was used to exclude any mental disorders;
④ PSQI score < 7;
⑤ Understand the content of the study and sign the informed consent form.

排除标准：

① 既往和目前有中枢神经系统疾病及其他严重躯体疾病； ② 酒精、药物或其他精神活性物质滥用者； ③ 药物依赖或精神分裂症引起的继发性抑郁发作； ④ MRI 禁忌症。

Exclusion criteria：

① Pre-existing and current central nervous system disorders and other serious physical illnesses; ② Alcohol, drug or other psychoactive substance abusers; ③ Secondary depressive episodes due to drug dependence or schizophrenia; ③ Depressive episodes secondary to drug dependence or schizophrenia. ④ Contraindications to MRI.
