研究目的:

类风湿关节炎(RA)是一种高致残性自身免疫病。中性粒细胞NETosis释放NETs在RA发病中发挥重要作用,而MLKL可通过在中性粒细胞膜上打孔介导NETs释放至胞外,可能参与RA炎症与骨破坏,但机制未明。我们前期发现从MLKL基因敲除小鼠中分离出的中性粒细胞经体外刺激后发生NETosis释放NETs的能力明显减低。本研究拟利用体内外实验探讨通过MLKL抑制中性粒细胞NETosis在RA治疗中的作用,阐明中性粒细胞NETosis在RA中的病理机制,为RA治疗提供新方法和新靶点。  

Objectives of Study:

Rheumatoid arthritis (RA) is a highly disabling autoimmune disease. The release of NETs from neutrophils plays an important role in the pathogenesis of RA, and MLKL can mediate the release of NETs to the extracellular space by punching holes in the neutrophil membrane, which may be involved in RA inflammation and bone destruction, but the mechanism is unclear. We previously found that neutrophils isolated from MLKL gene knockout mice had significantly reduced ability to release NETosis after in vitro stimulation. This study aims to explore the role of MLKL in inhibiting neutrophil NETosis in RA treatment through in vitro and in vivo experiments, elucidate the pathological mechanism of neutrophil NETosis in RA, and provide new methods and targets for RA treatment.

纳入标准:

Inclusion criteria

排除标准:

1.RA患者排除标准1:血液系统、肝、肾功能异常:入组前4周内WBC<4*109/L, PLT<100*109/L;ALT和/或AST >40U/L; 肌酐清除率小于20 mL/min/1.73m2;
2.RA患者排除标准2:有严重不能控制的高血压者;
3.RA患者排除标准3:妊娠或哺乳女性;
4.RA患者排除标准4:肿瘤;
5.RA患者排除标准5:急慢性感染性疾病,如乙肝、活动性结核等;
6.正常对照排除标准:合并任何自身免疫性疾病的体检者;

Exclusion criteria:

1.Exclusion criteria for RA patients 1:Abnormal blood system, liver, and kidney function: within 4 weeks before enrollment, WBC<4*109/L, PLT<100*109/L; ALT and/or AST>40U/L; Creatinine clearance rate less than 20 mL/min/1.73m2;
2.Exclusion criteria for RA patients 2:Individuals with severe uncontrolled hypertension;
3.Exclusion criteria for RA patients 3:Pregnant or lactating women;
4.Exclusion criteria for RA patients 4:tumor;
5.Exclusion criteria for RA patients 5:Acute and chronic infectious diseases, such as hepatitis B, active tuberculosis, etc;
6.Normal control exclusion criteria: Combination of any autoimmune diseases;
