研究目的：

①探讨针刺改善 KOA 患者大脑网络的异常功能连接模式，明确针刺改善 KOA 慢性疼痛发生的可能中枢靶点。 ②明确针刺改善 KOA 慢性疼痛的长期中枢效应机制。

Objectives of Study：

① Exploring the improvement of abnormal functional connectivity patterns in the brain network of KOA patients through acupuncture, and identifying potential central targets for acupuncture to improve chronic pain in KOA. ② Clarify the long-term central effect mechanism of acupuncture in improving chronic pain in KOA.

药物成份或治疗方案详述：

纳入标准：

Inclusion criteria

排除标准：

（1）有膝关节手术史或正在等待膝关节手术（膝关节置换或膝关节镜手术）；（2）评价关节 1 年之内有关节镜检查史、近 6 个月有关节腔注射史；（3）其他疾病引起的膝部疼痛（如关节游离体、关节腔严重积液、感染、恶性肿瘤、自身免疫疾病、外伤、骨折、痛风等）；（4）严重的急性或慢性器质性或精神类疾病；（5）凝血功能障碍疾病（如血友病等）；（6）备孕、妊娠期及哺乳期妇女；（7）幽闭恐惧症者或体内携带心脏起搏器、金属制品等其他MRI 检查禁忌者；（8）MRI 扫描中发现有明确器质性病变或严重头部解剖结构不对称者；（9）近 1 月内接受推拿或针灸治疗；（10）对针具和酒精过敏或者恐惧扎针者；（11）近 3 个月内参加其他临床研究者。

Exclusion criteria：

(1) Having a history of knee surgery or waiting for knee surgery (knee replacement or knee arthroscopic surgery); (2) Evaluate a history of arthroscopy examination within 1 year and a history of intra-articular injection in the past 6 months for the joint; (3) Knee pain caused by other diseases (such as joint dissociation, severe joint effusion, infection, malignant tumors, autoimmune diseases, trauma, fractures, gout, etc.); (4) Severe acute or chronic organic or psychiatric diseases; (5) Coagulation dysfunction diseases (such as hemophilia); (6) Pregnant, pregnant, and lactating women; (7) Individuals with claustrophobia or those who carry pacemakers, metal products, or other contraindications for MRI examination; (8) Those with clear organic lesions or severe head anatomical asymmetry found in MRI scans; (9) Received massage or acupuncture and moxibustion treatment in the past 1 month; (10) Individuals who are allergic to needles and alcohol, or are afraid of puncturing; (11) Participate in other clinical researchers within the past 3 months.

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：	临床筛选及评价数据采用纸质版病例报告表（CRF）。描述评估和收集结局指标、基线和其他试验数据由相关研究人员及时记录于 CRF 表，以备查询，若有修改，严格按照 CRF 表的修改说明进行详细标记。 fMRI 数据通过 DVD 光盘进行刻录，研究者需要及时进行数据格式的转化，查看影像数据是否完整以及检查相应的头动信息等。
Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：	Clinical screening and evaluation data were collected and recorded on paper case report forms (CRFs). The assessment and collection of outcomes, baseline data, and other trial data were recorded by relevant researchers on the CRF forms in a timely manner for future reference. Any modifications to the CRF forms were marked with detailed notes according to the modification instructions. fMRI data were recorded on DVD discs, and researchers were required to convert the data format in a timely manner to view the imaging data and check the corresponding head movement information.
