研究目的：

1.设计内镜下新的肠道菌群取样方法。 2.基于16SrDNA基因扩增子技术，进一步阐述MAFLD脂肪肝患者和健康人员的菌群特征及差异。 3.基于菌群结果，分析其与胆汁酸-FxR调节宿主脂质代谢和胰岛素抵抗的潜在机制。  

Objectives of Study：

1. Design a new sampling method by endoscopy. 2. Based on the 16S rDNA gene amplicon technology, the characteristics of MAFLD patient's microbiota were further elaborated. 3. Based on the results of microbiota, the potential mechanism of its regulation of host lipid metabolism and insulin resistance with bile acid-FxR was analyzed.

纳入标准：

Inclusion criteria

排除标准：

健康人群组排除标准： （1）不符合纳入标准者； （2）年龄<18岁或>60岁者； （3）入组前一个月内使用了抗生素且不愿停药洗脱；目前服用微生态制剂、胃肠动力药等可影响肠道菌群的制剂患者； （4）存在fibrotouch、肠镜检查禁忌症； （5）精神或法律上的残疾患者； （6）育龄期妇女且研究期间准备怀孕患者； （7）合并有重要脏器(如心、脑、肺、肝、肾)严重疾病者； （8）患者3个月内参加了其他临床试验； （9）研究者认为不适合入组的其他患者。 脂肪肝组排除标准： （1）不符合纳入标准者； （2）年龄<18岁或>60岁者； （3）饮酒史＞5年，折合乙醇量男性≥40g/d，女性 ≥ 20g/d; 或2周内有大量饮酒史，折合乙醇量 ＞80g/d; （4）病毒性肝炎、药物性肝病、遗传因素等其他可导致脂肪肝的特定疾病; （5）入组前一个月内使用了抗生素且不愿停药洗脱；目前服用微生态制剂、胃肠动力药等可影响肠道菌群的制剂患者； （6）存在fibrotouch、肠镜检查禁忌症； （7）精神或法律上的残疾患者； （8）育龄期妇女且研究期间准备怀孕患者； （9）合并有重要脏器(如心、脑、肺、肝、肾)严重疾病者； （10）患者3个月内参加了其他临床试验； （11）研究者认为不适合入组的其他患者。

Exclusion criteria：

Exclusion criteria for healthy population group: (1) Those who do not meet the inclusion criteria; (2) Individuals under the age of 18 or over 60 years old; (3) Used antibiotics within one month prior to enrollment and refused to discontinue medication for elution; Currently, patients taking probiotics, gastrointestinal motility drugs, and other preparations that can affect gut microbiota; (4) There are contraindications for fibrotouch and colonoscopy examination; (5) Patients with mental or legal disabilities; (6) Women of childbearing age who are preparing to become pregnant during the study period; (7) Patients with severe diseases of important organs such as heart, brain, lungs, liver, and kidneys; (8) The patient participated in other clinical trials within 3 months; (9) Researchers believe that other patients are not suitable for inclusion. Exclusion criteria for fatty liver group: (1) Those who do not meet the inclusion criteria; (2) Individuals under the age of 18 or over 60 years old; (3) Alcohol consumption history>5 years, equivalent to ethanol intake ≥ 40g/d for males and ≥ 20g/d for females; Or have a history of heavy alcohol consumption within 2 weeks, equivalent to an ethanol level greater than 80g/d; (4) Specific diseases that can lead to fatty liver, such as viral hepatitis, drug-induced liver disease, genetic factors, etc; (5) Used antibiotics within one month prior to enrollment and refused to discontinue medication for elution; Currently, patients taking probiotics, gastrointestinal motility drugs, and other preparations that can affect gut microbiota; (6) There are contraindications for fibrotouch and colonoscopy examination; (7) Patients with mental or legal disabilities; (8) Women of childbearing age who are preparing to become pregnant during the study period; (9) Patients with severe diseases of important organs such as heart, brain, lungs, liver, and kidneys; (10) The patient participated in other clinical trials within 3 months; (11) Researchers believe that other patients are not suitable for inclusion.
