研究目的:

本项目将骨质疏松早期诊断相关的分子标志物和血浆Hcy进行联合检测,有利于构建用以在高危人群中进行大规模筛查的早期预警模型,提高骨质疏松发病预测的准确性。本项目拟计划结合基因表达综合数据库(GEO)中的甲基化芯片数据和表达谱芯片数据,筛选出差异表达的基因且具有潜在调控功能的甲基化位点;选择出易感SNP位点进行基因分型,寻找与骨质疏松症易感性相关的候选位点;对样本血浆中的高同型半胱氨酸水平(tHCY)进行检测,用以联合两类分子标志物,建立一个用于骨质疏松早期诊断的预警模型。后期的分子机制研究为制定新的骨质疏松症预防、治疗策略提供实验依据。  

Objectives of Study:

This project will be related to the molecular markers and plasma Hcy combined detection in early diagnosis of osteoporosis, is conducive to building early warning model to mass screening in the high risk population, to improve the accuracy of predicting the onset of osteoporosis. This project is planned to be combined with gene expression microarray data integrated database (GEO) and expression of methylation microarray data in the spectrum, screened the methylation sites of differentially expressed genes and potential regulatory functions; selection of susceptible SNP loci were genotyped for osteoporosis susceptibility candidate loci related to; high levels of homocysteine in plasma samples (tHCY) were detected with combined two kinds of molecular markers, establish an early warning model for early diagnosis of osteoporosis. The molecular mechanism of the later study provides experimental basis for the development of new osteoporosis prevention and treatment strategies.

纳入标准:

Inclusion criteria

排除标准:

①合并糖尿病、甲状腺功能亢进、甲状旁腺功能亢进等影响骨代谢的内分泌疾病者;
②合并肝肾功能异常、胃炎、肠炎、类风湿性关节炎或骨肿瘤等恶性肿瘤者。
③半年内曾使用过激素或其他影响骨代谢的药物者;
④一年内发生骨折等导致肢体运动功能受限者。

Exclusion criteria:

(1) endocrine disorders such as diabetes mellitus, hyperthyroidism, hyperparathyroidism and so on affecting bone metabolism;
(2) patients with malignant liver and kidney dysfunction, gastritis, enteritis, rheumatoid arthritis or bone tumors;
(3) the use of hormones or other drugs that affect bone metabolism in half a year;
(4) fractures within one year lead to limb motor dysfunction.
