研究目的：

观察和评价“疏肝调神”法针刺干预失眠患者睡眠质量、改善其异常情绪及日间疲劳的临床疗效。在此基础上，基于神经血管耦合机制，利用功能近红外光谱成像（functionality near infrared spectroscopy，fNIRs）技术，检测失眠患者前额叶和顶叶血氧浓度变化，筛选失眠异常响应的关键脑区。同时探讨“疏肝调神” 法针刺对失眠患者额顶网络功能损害的影响和调节作用，初步阐明针刺干预失眠的脑功能机制，以期为失眠的针灸效应机制研究和临床应用提供依据。  

Objectives of Study：

To observe and evaluate the clinical efficacy of the acupuncture method of "dredging the liver and regulating the spirit" in intervening the sleep quality of insomnia patients and improving their abnormal mood and daytime fatigue. Based on the neurovascular coupling mechanism, fNIRs technology was used to detect the changes of blood oxygen concentration in the prefrontal and parietal lobes of insomnia patients, and to screen the key brain regions of insomnia response. At the same time, to explore the effects of acupuncture on the frontal-parietal network function of insomnia patients, and initially elucidated the cerebral function mechanism of acupuncture intervention in insomnia, with a view to providing a basis for the study of the effect mechanism of acupuncture on insomnia and its clinical application.

纳入标准：

Inclusion criteria

排除标准：

1.合并有严重的基础疾病，如恶性肿瘤、心肝肾功能不全、全身营养不良及风湿免疫疾病等； 2.药物或酒精所致失眠者； 3.合并有严重的抑郁、焦虑、双相情感障碍等精神类疾病； 4.对艾司唑仑不能耐受； 5.同期参加其他临床试验； 6.有自发性出血倾向或损伤后出血不止的病症（血小板减少性紫癜、白血病、血友病等）及局部皮肤感染不宜实施针刺治疗； 7.患者不愿意或者无法按要求配合完成试验。

Exclusion criteria：

1. Concomitant with severe underlying diseases, such as malignant tumors, heart, liver, and kidney dysfunction, systemic malnutrition, and rheumatic immune diseases; 2. Insomnia caused by drugs or alcohol; 3. Concomitant mental illnesses such as severe depression, anxiety, and bipolar disorder; 4. Inability to tolerate estazolam; 5. Participate in other clinical trials at the same time; 6. Acupuncture treatment is not recommended for symptoms with spontaneous bleeding tendencies or persistent bleeding after injury (such as thrombocytopenic purpura, leukemia, hemophilia, etc.) and local skin infections; 7. Patients are unwilling or unable to cooperate in completing the experiment as required.

本研究采用完全随机设计。借助SPSS26.0软件下的随机数生成器，将纳入本研究的122例失眠患者，按照就诊顺序依次编号1-122，产生122个对应的随机数，进一步对随机数编秩，得到另一列随机数，最后对第二次得到的随机数进行秩次排序，规定秩次1-61为西药组，62-122为针刺组。考虑到受不可控因素的影响，整个研究时间较短，为保证两组纳入的受试者均衡，以10个为一组，按上述操作完成随机分组。

Randomization Procedure (please state who generates the random number sequence and by what method)：

A completely randomised design was used in this study. With the help of random number generator under SPSS26.0 software, 122 insomnia patients included in this study were sequentially numbered 1-122 according to the order of consultation to generate 122 corresponding random numbers, further ranked the random numbers to obtain another column of random numbers, and finally ranked the random numbers obtained for the second time, stipulating that the rank 1-61 was the western medicine group, and 62-122 was the acupuncture group. Considering the influence of uncontrollable factors and the short time of the whole study, in order to ensure a balanced number of subjects included in the two groups, the random grouping was completed in groups of 10 as described above.

盲法：

由于针刺操作的特殊性，本研究无法实现双盲设计。为提高研究质量，本研究由第三方对受试者进行随机分组。此外，进行针刺操作者，不参与量表评定和数据处理。

Blinding：

A double-blind design could not be achieved in this study due to the specificity of the needling operation. To improve the quality of the study, subjects were randomised by a third party. In addition, those who performed the needling operation were not involved in scale assessment and data processing.
