评估以携带乙型肝炎病毒抗原基因的重组腺相关病毒感染自体树突状细胞诱导特异性细胞毒性T淋巴细胞治疗e抗原阳性慢性乙型肝炎的安全性和e抗原及S抗原转阴或转换的疗效  

Objectives of Study：

To Assesse the safety and preliminary efficacy of treatment with specific cytotoxic T lymphocytes (CTLs) induced by autologous dendritic cells(DCs) transduced with adeno-associated virus(AAV) carrying different hepatitis B virus (HBV) antigen gene fragments(S, C, E, X) in chronic hepatitis B（CHB）patients

Description for medicine or protocol of treatment in detail：

 

纳入标准：

Inclusion criteria

排除标准：

（1）合并其它肝炎病毒和HIV感染；（2）肝硬化失代偿；（3）自身免疫性疾病；（4）肝癌及其他恶性肿瘤； （5）肝功明显异常：总胆红素>2倍正常值上限(ULN)、ALT或AST>10×ULN；（6）过敏体质；（7）其它严重心脑肺肾和内分泌疾病如甲状腺功能异常；（8）T细胞淋巴瘤患者; （9）2周内有严重感染患者; （10）对粒细胞集落刺激因子等生物制品过敏的患者;（11）正在进行全身放疗、化疗和免疫抑制剂的患者;（12）血常规明显异常：白细胞总数低于3.5x10的9次方，血红蛋白低于110g/L；（13）怀孕或哺乳期妇女；（14）精神病或有精神病家族史；(15)吸毒或酗酒；（16）其它已知的原发性或继发性肝脏损伤，如酒精性肝炎、药物性肝炎、自身免疫性肝炎、血色病、α-1抗胰蛋白酶缺乏症、Wilson氏病、Gilbert综合征和Dubin-John综合征等先天性或代谢性疾病以及经治医师认为不适合纳入的患者。

Exclusion criteria：

1. Co-infected with other hepatitis virus (including HCV, HAV, HDV, HEV) or HIV
2. Decompensated liver disease
3. autoimmune diseases
4. ALT or AST> 10 ULN, or total bilirubin > 2 mg/dL
5. Hepatocellular carcinoma (HCC) or other malignant tumors;
6. T cell lymphoma;
7. Severe infection within past 2 weeks;
8. Allergy to biological agents such as granulocyte colony-stimulating factor;
9. Undergoing systemic radiotherapychemotherapy or immunosuppressive agents;
10. Blood routine was significantly abnormal: the total number of white blood cells was lower than 3.5x10^9, and the hemoglobin was less than 110g/L;
11. Subjects or his/her family members with a history of seizures or psychiatric disability
12. Clinically significant renal, respiratory, cerebral, cardiovascular or endocrine disease such as?severe thyroid dysfunction
13. Pregnancy and lactation
14. Voluntarily withdrew from the study;
15. Drug addicts or alcoholism
16. One or more additional known primary or secondary causes of liver disease, e.g., alcohol-induced hepatitisdrug-induced hepatitisautoimmune hepatitishemochromatosisalpha-1 antitrypsin deficiency, Wilson's Disease, Gilbert's syndrome, Dubin-Johnson syndrome and other reasons considered to be exclused by the investigator.
