研究目的:

脊髓损伤(Spinal Cord Injury,SCI)是一种严重的中枢神经系统损伤,具有高发病 率、高致残率、高死亡率等特点,目前临床上尚缺乏有效的干预方法。经颅直流电刺激(Tran scranial direct current stimulation,tDCS)作为一种非侵入性脑刺激技术,已被证明 可改善多种原因所致神经系统损伤。项目组前期研究证明tDCS可抑制神经元凋亡、促进细胞自 噬、减少脊髓继发性病理损伤,从而对SCI大鼠起保护作用;前期临床研究发现tDCS可促进SCI 患者运动功能康复,但其具体机制值得深入研究。本项目在前期研究基础上,基于代谢组学探 究SCI患者tDCS干预前后差异代谢产物的时间依赖性变化,及其与功能学评估的相关性,筛选 特异性生物标志物。本研究成果将为tDCS通过调节代谢网络促进SCI运动功能障碍的康复机制 提供一定理论依据,也为SCI临床治疗提供新的方向和策略。  

Objectives of Study:

Spinal cord injury (SCI) is a serious injury of central nervous system with high morbidity, disability and mortality, for which there is no effective intervention method in clinic at present. Transcranial direct current stimulation (tDCS), as a non-invasive brain stimulation technique, has been proved to improve the nervous system injury due to a variety of causes. The preliminary study of this project team proved that tDCS could inhibit neuronal apoptosis, promote autophagy and reduce secondary pathological injury of spinal cord, thus playing a protective role in SCI rats. Previous clinical studies have found that tDCS can promote the recovery of motor function in SCI patients, and its specific mechanism is worthy of further study. On the basis of previous studies, this project explored the time-dependent changes of differential metabolites in SCI patients before and after tDCS intervention based on metabonomics, their correlation with functional evaluation and screened specific biomarkers. The research results will provide a theoretical basis for tDCS to promote the rehabilitation mechanism of SCI motor dysfunction by regulating metabolic network, and provide a new direction and strategy for clinical treatment of SCI.

排除标准:

伴严重脑、心、肝、肾、肺等重要脏器疾病者,有严重意识、认知障碍或精神病患者,合并严重头部外伤者以及体内有植入式电子设备(如心脏起搏器等) 不能配合检查与治疗者。

Exclusion criteria:

Patients with severe brain, heart, liver, kidney, lung and other important organ diseases, patients with severe consciousness, cognitive impairment or mental illness, patients with severe head trauma, and implanted electronic devices (such as pacemakers, etc.) ) cannot cooperate with examination and treatment.
