研究目的：

（1）建立临床预测模型；（2）运用分子生物学、免疫学等方法，建立分子相互作用和调控网络，明确遗传/表观遗传以及微生态参导致免疫耐受失衡在疾病发生发展中的作用机制及靶点；（3）通过多维度数据分析，建立不同临床表型老年 AID 受累器官的生物标志物图谱;将其结合临床特征，形成早期诊断、疗效预测和预后模型。

Objectives of Study：

(1)To establish a clinical prediction model; (2)Using molecular biology and immunology to establish molecular interactions and regulatory networks to clarify the mechanisms and targets of genetic/epigenetic and microecological parameters leading to imbalance of immune tolerance in disease development; (3)To establish biomarker profiles for different clinical phenotypes of aged AID-involved organs through multidimensional data analysis; combine them with clinical features to form early diagnosis, efficacy prediction and prognosis models.

Description for medicine or protocol of treatment in detail：

纳入标准：

Inclusion criteria

(1)Age: 18-65 years, patients with SLE or RA;
(2)The diagnosis of SLE patients should meet the 1987 revised American College of Rheumatology classification criteria; disease activity was assessed using the systemic lupus erythematosus disease activity index (SLEDAI) scoring system; SLEDA score >= 5 , ANA antibody >= 1:40; hormone maintenance dose <= 3 tablets/day for three months and no biologics/immunosuppressive agents for six months;
(3)The diagnosis of RA patients all need to meet the diagnostic criteria of rheumatoid arthritis classification criteria and scoring system (ACR/EULAR, 2010) classification; disease activity was assessed using 28 joint disease activity (DAS28) score; 4 swollen and 4 painful joints, seropositive, erythrocyte sedimentation rate (ESR) >28 mm/h; methotrexate or methotrexate + leflunomide treatment was ineffective for 3 months, and no other anti-rheumatic drugs (available low-dose hormones ± NSAIDs) were used in the last 3 months;
(4)All patients have not been treated with glucocorticoids, immunosuppressants, disease-modifying antirheumatic drugs or biologics within 6 months;
(5) The healthy control population should be sex- and age-matched to SLE or RA patients, and the enrolled population should have no history of SLE, RA or other AIDs.

排除标准：

(1)近1月服用抗生素、抗惊厥药物或患有胃肠道型感冒等影响肠道菌群的患者或健康对照；
(2)近期患有感染性疾病、恶性肿瘤、白血病、急性心衰、急性冠脉综合征等严重基础疾病的患者或对照；
(3)有严重的、进展性的或未控制的肾脏、肝脏、血液、胃肠、内分泌、肺部、心脏、神经、精神或脑部疾病的症状或体征；
(4)行器官移植手术。

Exclusion criteria：

(1)Patients or healthy controls taking antibiotics, anticonvulsant medications or suffering from gastrointestinal-type colds that affect the intestinal flora in the last month;
(2)Patients or healthy controls with recent serious underlying diseases such as infectious diseases, malignancies, leukemia, acute heart failure, acute coronary syndrome;
(3)Patients with signs or symptoms of severe, progressive or uncontrolled renal, hepatic, hematologic, gastrointestinal, endocrine, pulmonary, cardiac, neurologic, psychiatric or cerebral diseases;
(4)Patients who have undergone organ transplantation.
