研究目的：

1. 解析湿疹患儿皮肤微生物组的结构特征以及可能的病因机制； 2. 分离重要的皮肤微生物组成员（细菌），构建体外皮肤菌群模型； 3. 应用皮肤菌群模型，评价2种有开发潜能的功能成分或因子对皮肤菌群的调节作用。

Objectives of Study：

1. To analyze the structural characteristics of skin microbiome in children with eczema and the possible etiological mechanism; 2. Isolate important skin microbiome members (bacteria) and construct in vitro skin microbiota model; 3. The skin flora model was used to evaluate the regulatory effect of two potential functional components or factors on the skin flora.

纳入标准：

Inclusion criteria

排除标准：

1.有严重的未控制的慢性肝、肾、心脏、血液系统、内分泌及肺部疾病或先天性疾病；
2.有恶性肿瘤史；
3.罹患自身免疫性疾病或免疫缺陷性疾病；
4.2周内有抗生素、抗真菌药及糖皮质激素用药史；
5.有近2日洗浴史。

Exclusion criteria：

1. Serious uncontrolled chronic diseases of liver, kidney, heart, blood system, endocrine and lung or congenital diseases;
2. History of malignant tumor;
3. Suffering from autoimmune or immunodeficiency diseases;
4. History of antibiotic, antifungal and glucocorticoid use within 2 weeks;
5. Have taken baths for the past two days.

本研究是一项开放性、对照、单中心研究。拟分两组，婴儿湿疹组与健康婴儿组，各50例。本研究招募受试者主要来自门诊诊断为湿疹的患者及非湿疹的健康受试者。提取样品微生物组DNA，其中皮肤样本数量AD组200，健康组100。用PCR扩增16S rRNA基因V3-V4区，利用Illumina Miseq测序技术分析菌群的组成，采用主成分分析（PCA）、偏最小二乘法判别分析（PLS-DA）以及随机森林等方法进行数据分类模型的建立和检验，解析湿疹菌群的组成特征、寻找关键细菌和细菌群。选择AD组及健康组各20例典型样本，进行鸟枪法元基因组测序（Illumina Hiseq XTen平台），分析皮肤系统中微生物的结构和潜在功能。
