研究目的:

肠道菌群改变是促进脓毒症发生发展的重要机制之一,并对脓毒症的预后产生不良影响,研究发现其可能通过调节免疫功能影响患者的预后,但具体机制未完全阐明。在肿瘤领域的研究已经证明了肠道菌群变化对肿瘤患者抗PD-1及PD-L1的治疗疗效的影响,我们先前研究表明调节性T细胞(Treg细胞) 中的 PD-1 的表达对脓毒症患者的预后具有重要价值。本研究旨在评估脓毒症患者的肠道菌群及其代谢产物变化、肠道黏膜通透性改变,进一步分析其与Treg细胞共信号分子的表达、病情严重程度及预后的关系,为阐明脓毒症患者肠道菌群改变影响免疫功能的机制提供实验依据。  

Objectives of Study:

The change of intestinal flora is one of the important mechanisms to promote the occurrence and development of sepsis, and has a negative impact on the prognosis of sepsis. Studies have found that it may affect the prognosis of patients by regulating immune function, but the specific mechanism has not been fully clarified. Studies in the field of oncology have demonstrated the effect of intestinal flora changes on the efficacy of anti-PD-1 and PD-L1 therapy in tumor patients. our previous studies have shown that the expression of PD-1 in regulatory T cells (Treg cells) plays an important role in the prognosis of patients with sepsis. The purpose of this study was to evaluate the changes of intestinal flora and its metabolites and intestinal mucosal permeability in patients with sepsis, and to further analyze the relationship between intestinal flora and Treg cell co-signal molecule expression, disease severity and prognosis, so as to provide experimental basis for elucidating the mechanism of intestinal flora changes affecting immune function in patients with sepsis.

纳入标准:

Inclusion criteria

排除标准:

(1)年龄<18岁;
(2)各种原因无法排便患者;
(3)恶性肿瘤等慢性疾病终末期,预计生存时间<28天;
(4)存在自身免疫系统疾病、免疫缺陷或长期服用免疫抑制剂的患者;
(5)孕妇;
(6)患者及家属不同意参加本试验。

Exclusion criteria:

1. Patients less than 18 years old; 2. Patients who are unable to defecate for various reasons; 3. End-stage chronic diseases such as malignant tumors, the estimated survival time is less than 28 days; 4. Patients with autoimmune diseases, immune deficiencies, or long-term use of immunosuppressants; 5. Pregnant women; 6. Patients and their families do not agree to participate in this trial.
