本研究通过研究泊沙康唑口服混悬液在儿童体内的药动学,以及与环孢素的药物相互作用,并应用生理药动学原理、结合药物及其制剂性质,以及患者生理、病理状态,建立儿童患者口服泊沙康唑的药动学计算机仿真模型,实现: 1. 对泊沙康唑口服混悬液儿童体内药动学行为的定量预测,并应用于治疗药物监测,指导个体化给药; 2. 预测儿童患者组织器官的药物暴露程度,确保靶器官达到有效治疗浓度和疗效、降低非靶器官的毒副作用; 3. 预测与环孢素的药物相互作用,指导剂量调整。为开展无创治疗药物监测、减少采血频次提供依据。 |

Objectives of Study: |
This study intends to study the pharmacokinetic behavior of posaconazole oral suspension in children and its interactions with cyclosporine, then to use physiologically based pharmacokinetic principles, combined with drugs properties, as well as the patient's physiological and pathological state, to establish a computer simulation model of posaconazole in children: 1. To predict the pharmacokinetic behavior of posaconazole oral suspension in children and its application in therapeutic drug monitoring to guide individual drug administration; 2. To predict the drug exposure in the tissues and organs to ensure that the target organs achieve effective therapeutic concentration and efficacy, and reduce the toxic and side effects of non-target organs; 3. To predict drug interactions with cyclosporine and guide dose adjustment. And provide basis for developing of non-invasive therapeutic drug monitoring and reducing blood collection frequency. |
