对比研究急性高强度间歇运动(HIIT)和传统中等强度运动(MICT)对T2D患者糖脂代谢和血浆外泌体miRNAs的影响,通过对差异表达的血浆外泌体miRNAs进行定量检测,将筛选得到的差异miRNA与糖脂代谢相关指标进行相关性分析,寻找运动改善T2D的新靶标,进一步对差异表达的外泌体miRNA应用生物信息学分析,探讨运动对T2D患者血浆外泌体的调控机制。以期为运动防治T2D的基础研究及实践应用提供新方向。也为老龄化加速背景下运动促进健康与慢病防治的机制与应用提供依据。  

Objectives of Study:

To explore the effects of acute high-intensity intermittent exercise (HIIT) and traditional moderate intensity exercise (MICT) on glucose and lipid metabolism and plasma exosomal miRNsA in T2D patients, and through the quantitative detection of differentially expressed plasma exosomal miRNAs, the correlation between the screened differential miRNAs and glucose and lipid metabolism related indicators will be analyzed. New targets for exercise to improve T2D will be found. Further, bioinformatics analysis will be applied to the differentially expressed exosomal miRNA to explore the regulatory mechanism of exercise on plasma exosomes in T2D patients.

纳入标准:

Inclusion criteria

排除标准:

1.空腹血糖>11.1 mmol/L;HbA1c>8.0%;BMI>28或<18.5 kg/m2;甘油三酯>5.7 mmol/L;
2.冠脉CT或造影检查显示血管狭窄大于40%,或存在严重心血管疾病及风险者;
3.肝功能、甲状腺功能、肌酸激酶(CK)异常者;
4.胰岛素使用者;
5.吸烟者;
6.收缩压≥160mmHg,舒张压>90mmHg或有低血压(静息坐位血压<90/50mmHg)患者;
7.各种运动禁忌症患者;
8.起搏器使用者;
9.实验前3个月参加其他实验者;
10.实验前6个月内参与了规律的体育锻炼者。

Exclusion criteria:

1. Fasting blood glucose > 11.1 mmol/L; HbA1c > 8.0%; BMI > 28 or < 18.5 kg/m2; triglyceride > 5.7 mmol/L;
2. Coronary CT or angiography shows that the vascular stenosis is greater than 40%, or there are serious cardiovascular diseases and risks;
3. Abnormal liver function, thyroid function and creatine kinase (CK);
4. Insulin users;
5. Smokers;
6. Systolic blood pressure >= 160mmHg, diastolic blood pressure > 90mmHg or patients with hypotension (resting sitting blood pressure < 90/50mmHg);
7. Patients with various exercise contraindications;
8. Pacemaker users;
9. Participants who participated in other experiments 3 months before the experiment;
10. Those who participated in regular physical exercise within 6 months before the experiment.
