研究目的:

1.从多组学角度分析正常人群和2型糖尿病人群药物干预后的多组学产物及肠道菌群的变化,加深对现有药物作用机制理解。 2.从代谢组学和肠道菌群角度进一步理解糖尿病发病机制,寻找糖尿病及糖尿病并发症可能的早期代谢标志物,为指导糖尿病的早期诊断和个体化管理提供研究证据。  

Objectives of Study:

1. From the perspective of multiomics, the changes of multiomics and intestinal flora in normal people and type 2 diabetes population after drug intervention were analyzed, so as to deepen the understanding of the mechanism of action of existing drugs. 2. To further understand the pathogenesis of diabetes from the perspective of metabonomics and intestinal flora, and to search for possible early metabolic markers of diabetes and diabetes complications, so as to provide research evidence for the early diagnosis and individualized management of diabetes.

纳入标准:

Inclusion criteria

排除标准:

1. 妊娠状态; 2. 有未控制的甲亢、甲减、库欣病等其他影响糖代谢的内分泌疾病; 3. 有未控制的类风湿性关节炎、红斑狼疮等免疫系统性疾病; 4. 有肝功异常(ALT、AST 为正常上限 2.5 倍以上者)或肾功损害(血肌酐≥133umol/或者估计肾小球率过滤(eGFR)小于 30mL/min/1.73m2)或其他脏器功能不全者; 5. 有肿瘤病史,预估生存期小于五年者; 6. 近期有严重急性感染、手术、创伤事件; 7. 嗜酒及药物成瘾者; 8. 年龄小于18岁或大于75岁。

Exclusion criteria:

1. Pregnancy status; 2. Uncontrolled hyperthyroidism, hypothyroidism, Cushing's disease and other endocrine diseases that affect glucose metabolism; 3. Uncontrolled rheumatoid arthritis, lupus erythematosus and other immune systemic diseases; 4. Abnormal liver function (ALT and AST are more than 2.5 times the upper limit of normal) or impaired kidney function (serum creatinine) >= 133 umol/L or estimated glomerular rate filtration (eGFR) less than 30mL/min/1.73m2) or other organs are not fully functioning; 5. Patients with a history of tumor and an estimated survival of less than five years; 6. Severe acute infection, surgery and trauma have occurred recently; 7. Alcohol and drug addicts; 8. Under 18 years old or over 75 years old.

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

研究者根据受试者的原始观察记录,将数据及时、完整、正确、清晰地载入病例报告表,录入采用相应的数据库系统双人双机录入,之后对数据库进行两遍比对,电子数据文件分类保存,并有多个备份保存于不同磁盘或记录介质上,妥善保存,防止破坏。

According to the original observation records of the subjects, the researchers loaded the data into the case report form in a timely, complete, correct and clear manner, and input the data into the corresponding database system with two persons and two computers. After that, the database was compared twice, and the electronic data files were classified and saved, and multiple backups were saved on different disks or recording media, which were properly preserved to prevent damage.
