研究目的:

1.收集肥胖症及2 型糖尿病患者在减重代谢手术治疗时的胃周脂肪组织、空肠和十二指肠等组织样本,手术前后的血液、尿液及粪 便等标本,同时构建实验动物模型收集动物样本,建立 500-1000 人份生物样本库; 2.获得手术前后的肠道宏基因组学、蛋白质组学、脂质组学、代谢组学和转录组学等多层次组学数据; 3.阐明通过减 重代谢手术介导的肠道菌群调控糖脂代谢的作用机制,鉴定 2-3 个与其相关的糖脂代谢分子调控网络;确定 3-4 个关键调节分子和/或关键 调控通路; 4.探索调控糖脂代谢的新靶点,筛选治疗肥胖症和2型糖尿病糖脂代谢紊乱的新型的高效的调控药物,制备 1-2 个新型高效 糖脂代谢调控药物,并完成体内外实验验证。  

Objectives of Study:

1.Collect adipose tissue samples from patients during metabolic surgery, as well as blood, urine and stool samples before and after surgery. Meanwhile, build experimental animal models to collect animal samples, and establish a biobank of 500-1000 people; 2.Multi-level omics data including gut metagenomics, proteomics, lipidomics, metabolomics and transcriptomics were obtained before and after surgery; 3.Elucidate the mechanism of regulating glucose and lipid metabolism by gut microbiota, identify 2-3 related molecules which regulate the networks of glucose and lipid metabolism; Identify 3-4 key regulatory molecules and/or key regulatory pathways; 4.Explore new targets for the regulation of glucose and lipid metabolism, screen new and efficient regulatory drugs for the treatment of obesity and type 2 diabetes; prepare 1-2 new and efficient regulatory drugs for glucose and lipid metabolism and complete with experiments in vivo/in vitro.

纳入标准:

Inclusion criteria

排除标准:

1.发生严重糖尿病并发症的患者,如:腿部溃疡、严重肾损伤、严重神经疾病、严重视网膜病等;
2.过去3个月内使用过抗生素,并持续3天或以上;
3.酗酒(1周饮酒5次以上,平均每次2两白酒或半斤黄酒或5瓶啤酒以上);
4.过去6个月内患有严重的精神疾患;
5.过去3个月内因下列疾病接受药物治疗的:胆囊炎、消化道溃疡、尿路感染、急性肾盂肾炎、膀胱炎、甲亢等甲状腺功能异常;
6.进行过胃肠道手术,除阑尾炎、疝气手术外;
7.慢性、迁延性肝炎患者,肝硬化等严重肝脏疾病患者或乙肝病毒表面抗原(HBsAg)阳性并伴随有肝功能有异常者(血清谷丙转氨酶和谷草转氨酶是正常值的2.5倍);
8.患有垂体功能异常;严重器质性疾病如癌症、心肌梗塞以及中风等;肺结核和艾滋病等传染病;
9.过去3月内以任何药物方法(包括氟苯丙氨等食欲抑制剂类药物和甲状腺激素、孕酮、泻剂等和各种中药成分减肥药)或手术方式进行减肥者;
10.生活不能自理、不能清楚地回忆、回答问题者。

Exclusion criteria:

1.Patients with severe diabetes complications, such as leg ulcers, severe kidney injury, severe neurological disease, severe retinopathy, etc.;
2.Patients have taken antibiotics for 3 days or more in the past 3 months;
3.Alcoholism (drinking more than 5 times per week, with an average of more than 50 g of white wine or 500g of chinese rice wine or 5 bottles of beer each time);
4.Patients suffer from a serious mental disorder in the past 6 months;
5.Patients have thyroid dysfunction due to medical treatment for the following diseases in the past 3 months: cholecystitis, gastrointestinal ulcer, urinary tract infection, acute pyelonephritis, cystitis, hyperthyroidism, etc.;
6.Patients had undergone gastrointestinal surgery, except appendicitis and hernia surgery;
7.Patients with chronic and persistent hepatitis, cirrhosis and other serious liver diseases or hepatitis B virus surface antigen (HBsAg) positive accompanied by abnormal liver function (serum alanine aminotransferase and aspartate aminotransferase are 2.5 times of the normal value);
8.Patients suffer from abnormal pituitary function; Severe organic diseases such as cancer, myocardial infarction and stroke; Infectious diseases such as tuberculosis and HIV/AIDS;
9.Weight loss in the past 3 months by drug method (such as: fluorophenylpropanol, thyroid hormone, progesterone, laxatives and traditional Chinese medicine) or surgical methods;
10.Patients can not take care of themselves, can not clearly recall, answer questions.
