研究目的：

本研究基于中医传统理论“胃不和则卧不安”，围绕“脑-肠-菌”轴功能失衡这一失眠的重要发病机制，以针刺优势病种失眠为载体，以“和胃安神”为主要治法，提出“影响‘脑-肠-菌’轴GABA、Glu含量，调整中枢神经系统代谢和肠道菌群功能是‘和胃安神’针法治疗原发性失眠的重要机制”的研究假说，从关键靶分子GABA、Glu入手，首次联合磁共振波谱扫描、宏基因组学和高效液相色谱法等技术，从肠道菌群-肠道-脑的互动角度，阐释经典中医理论“胃不和则卧不安”和现代医学“脑-肠-菌”轴失调的相互关系，探讨针刺干预胃肠道功能治疗原发性失眠的可行性。该成果有望为针刺调节原发性失眠的机制研究提供新的思路和证据，也可为疾病药物治疗提供新的靶点。

Objectives of Study：

Based on the mechanism and the Traditional Chinese Medicine (TCM) classics of Stomach Disharmony Causes Sleeping Difficulties, our research first raised the prognosis of the core mechanism of Hei Wei An Shen (Harmonizing Stomach to Calm mind) needing therapy is interfering the content of GABA and Glu within the BIBS, as well as regulating the metabolism of central nervous system and functions of intestinal flora.We started from the target molecular of GABA and Glu, and combined with the utilizations of Magnetic Resonance Spectrum Scan, Metagenomics and High-Performance Liquid Chromatography for the first time in the history, interpreted correlations between the TCM theory of disharmony in stomach caused irritable sleep and BIBS, and further discussed the feasibility of regulating sleeping patterns by Harmonizing Stomach needling therapy.We expect this project to be a new proof of improving Primary insomnia by acupuncture, hence provides reliable evidence for future research and references our results to relevant pharmacotherapy in clinical practice.

纳入标准：

1.性别不限，18岁≤年龄≤65岁，右利手； 2.符合非器质性失眠的西医诊断标准； 3.符合中医脾胃不和型失眠的诊断； 4.匹兹堡睡眠质量指数（PSQI）评分＞7分； 5.患者签署知情同意书自愿参加本研究者。注：同时符合以上5项的患者，方可纳入本项研究。

Inclusion criteria

(1) Male or female; Aged 18 to 65 years old; dextral; (2) Meet the western medicine diagnostic criteria of non-organic insomnia( sleep latency over 30 minutes, and/or wake up at night more than 2 times per night, and/or the total sleep time<6 hours. Duration of those symptoms is more than 1 month); (3) Meet the diagnostic criteria of insomnia in TCM (Conform to the differentiation standard of disharmony between spleen and stomach syndrome); (4) Pittsburgh Sleep Quality Index (PSQI) score >7 points; (5) The patient signed the informed consent and voluntarily participated in the study.

排除标准：

1.合并有心脑血管、肝、肾和造血系统等严重危及生命的原发性疾病以及精神病患者、哮喘患者； 2.全身性疾病如疼痛、发热、咳嗽、手术等； 3.酗酒和（或）精神药物滥用和依赖（含安眠药）所致失眠者； 4.伴有明显的头痛、偏头痛，头部外伤史者； 5.过去1个月内服用抗生素，糖皮质激素，免疫抑制剂或中药的患者; 7.妊娠、准备妊娠或哺乳期妇女； 8.有磁共振扫描禁忌症者; 9.在1个月内参与其他的临床试验者；注：凡符合上述任何一条的患者，均予以排除。

Exclusion criteria：

(1) Patients with severe primary diseases such as cardiovascular, pulmonary, liver, kidney and hematopoietic systems; (2) With systemic diseases such as pain, fever, cough, surgery, etc.; (3) Insomnia caused by alcohol abuse and/or psychotropic substance abuse and dependence (including sleeping pills); (4) With obvious headache, migraine or history of head trauma; (5) Patients have taken antibiotics, glucocorticoids, immunosuppressive agents or Chinese medicine within last 1 month; (6) Patients have taken medicines treating insomnia at least 2 weeks before entering the study; (7) Pregnancy or lactation women; (8) With magnetic resonance scanning contraindications; (9) Participated in other clinical trials within 1 month.
