研究目的：

研究艾地苯醌在周期性麻痹治疗中的疗效和安全性。  

Objectives of Study：

To evaluate the efficacy and safety of Idebenone for Periodic Paralysis

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.基因确诊、临床确诊或临床很可能的高钾性或低钾性周期性麻痹患者；
2.男性或女性，≥18岁；
3.在治疗或未治疗情况下，受试者发作次数≥1次/周，但?3次/天；
4.促甲状腺素（TSH）和甲状腺素（T4）正常；
5.签署知情同意书；

Inclusion criteria

1. Genetically definite, clinically definite or clinically probable HypoKPP or HyperKPP as outlined in the protocol;
2. Male and female participants, age 18 and older who are able to comply with the study conditions;
3. Participants who have distinct regular episodes of weakness with an average frequency of > or = to 1 a week and < or = to 3 a day either on or off treatment, whichever is higher;
4. Normal thyroid-stimulating hormone (TSH) level and thyroxine(T4);
5. The patient or the patients legal representative have provided singed written informed consent.

排除标准：

1.Andersen-Tawil综合征：（满足以下3项中的任何1项)；
1）QT间期延长或室性心律失常
2）机体畸形（满足以下5项中的2项）
低位耳
身材矮小症
小颌畸形
先天性指（趾）弯曲
眼距过宽
3）KCNJ2基因突变
2.永久性肌无力；
3.对艾地苯醌过敏或产生不能耐受的副作用；
4. 合并肝、肾及活动性甲状腺疾病、限制性或阻塞性肺病、其他神经肌肉病或心脏疾病、存在各种原因造成的持续性腹泻或呕吐等顽固性离子丢失情况；
5.使用以下药物：利尿药、抗心律失常药物、皮质类固醇激素、β受体阻滞剂、钙离子通道拮抗剂、抗癫痫药物及镁剂；
6.怀孕；

Exclusion criteria：

1.Evidence for Andersen-Tawil syndrome (any one of the following 3 criteria):
(1) Prolonged QT interval or complex ventricular ectopy between attacks
(2) Distinctive physical features (2 of the following 5); Low set ears; Short stature; HyperKPPo-/micrognathia; Clinodactyly; HyperKPPo-/HyperKPPertelorism;
(3) KIR 2.1 gene mutation.
2. The patient has permanent weakness;
3. Allergy to Idebenone or intolerance to side effects;
3. Coincidental renal, hepatic, active thyroid disease, restrictive or obstructive lung disease, other neuromuscular disease, or heart disease;
4. Use of any of the following medications for reasons other than treatment of periodic paralysis: diuretics, antiarrhythmics, corticosteroids, beta-blockers, calcium channel blockers, antiepileptics, magnesium;
5. History of life-threatening episodes of respiratory muscle weakness or cardiac arrhythmias during attacks;
6. Pregnancy.
