研究目的：

胆囊癌是消化系统常见的恶性肿瘤之一，其恶性程度高，预后较差，手术切除是唯一可能治愈胆囊癌的治疗方式，早期胆囊癌有手术机会，预后相对较好，但中晚期胆囊癌患者往往预后不良，甚至失去手术机会，所以胆囊癌的早期诊断及其治疗至关重要。 生长分化因子15（growth differentiation factor 15，GDF15）是一种生长因子，属于TGF-β（转化生长因子β）超家族成员。相关研究表明，GDF15在正常生理情况下除胎盘外的组织中表达较少，但是在炎症、氧化应激、癌基因激活等病理情况刺激下，受累器官和血清中的GDF15表达均明显增高。目前已有学者在肝癌、胃癌、胰腺癌等多种癌症中发现，GDF15的高表达与肿瘤的复发、转移及患者的预后相关，但是GDF15与胆囊癌的相关性研究尚未见报道，本研究应用ELISA法检测胆囊癌、慢性胆囊炎、正常人血清中GDF15的表达，并用RT-PCR、免疫组化等技术检测GDF15在胆囊癌肿瘤组织、癌旁组织及正常组织中的表达情况，旨在发现GDF15在胆囊癌中的作用，可能会在一定程度上提高对胆囊癌的早期诊断的准确性,从而对胆囊癌的诊断及治疗提供指导作用。  

Objectives of Study：

Gallbladder cancer is one of the common malignant tumors of the digestive system. It has a high degree of malignancy and a poor prognosis. Surgical resection is the only treatment that can cure gallbladder cancer. There is a chance of surgery for early-stage gallbladder cancer, and the prognosis is relatively good, but patients with middle-advanced gallbladder cancer often have a poor prognosis and even lose the opportunity of operation, so the early diagnosis and treatment for gallbladder cancer are very important. Growth differentiation factor 15 (GDF15) is a growth factor that belongs to the TGF-β (transforming growth factor β) superfamily. Relevant studies have shown that GDF15 is less expressed in tissues other than the placenta under normal physiological conditions, but under the stimulation of inflammation, oxidative stress, oncogene activation and other pathological conditions, the expression of GDF15 in the affected organs and serum is significantly increased. At present, some scholars have found that the high expression of GDF15 is related to the recurrence, metastasis and prognosis of patients with liver cancer, gastric cancer and pancreatic cancer. However, the correlation between GDF15 and gallbladder cancer has not been reported. In this study, the expression of GDF15 in serum of gallbladder cancer, chronic cholecystitis and normal people was detected by ELISA,, and uses RT-PCR, immunohistochemistry and other techniques to detect the expression of GDF15 in gallbladder cancer tumor tissues, adjacent tissues and normal tissues, aiming to find the role of GDF15 in gallbladder cancer, may improve the accuracy of early diagnosis of gallbladder cancer to a certain extent, thereby providing guidance for the diagnosis and treatment for gallbladder cancer.

纳入标准：

1、原发性胆囊癌患者，术前均未行放疗、化疗、免疫治疗；
2、胆囊癌患者，均行胆囊癌根治术；
3、对照组随机选择与胆囊癌患者同期就诊的慢性胆囊炎患者和健康志愿者，均无基础疾病。

Inclusion criteria

1. Patients with primary gallbladder cancer, and all of them had not received radiotherapy, chemotherapy or immunotherapy before surgery;
2. All patients with gallbladder cancer underwent radical resection of gallbladder cancer;
3. In the control group, patients with chronic cholecystitis and healthy volunteers who were treated at the same time as patients with gallbladder cancer were randomly selected, and none of them had other basic diseases.
