研究目的：

建立治疗四类儿科疑难疾病（感染、免疫、移植、难治性病）的药物的DBS的HPLC-MS/MS检测方法；构建目标药物在儿科群体中的PK-PD-PG及PBPK模型；开发基于贝叶斯法的药物治疗达标预测工具。  

Objectives of Study：

To establish the dry blood spot (DBS) and high performance liquid chromatography and electrospray tandem mass spectrometry (HPLC-MS/MS) assay methods for drugs used to treat four types of pediatric complicated diseases (infection, autoimmune diseases, transplantation, refractory diseases); to develop PK-PD-PG and PBPK models of the drugs in pediatric population; to develop Bayesian estimators to predict the target attainment under different treatment regimens.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

（1）患有四类疾病中的一种：感染（新生儿/早产儿脓毒血症）、免疫（儿童系统性红斑狼疮、幼年性关节炎、血液肿瘤）、移植（骨髓移植、肝移植、肾移植）、慢性/难治病（难治性癫痫、难治性肾病综合征）；
（2）年龄为0～18岁；
（3）患病期间采用针对各类疾病的目标药物治疗；
（4）使用目标药物的特定给药方式

Inclusion criteria

(1) diagnosed as one of the four types of the diseases: infection (sepsis of newborn/premature), autoimmune diseases (systemic lupus erythematosus, juvenile arthritis, hematological malignancies);
(2) aged 0~18 years;
(3) used the specific drugs to treat the diseases;
(4) administered the drugs via the specific ways.

排除标准：

（1）有严重的出生缺陷；
（2）患儿或其母亲感染HIV或肝炎；
（3）重症患儿在研究期间无法存活的；
（4）对研究的目标药物过敏。

Exclusion criteria：

1. have severe inborn defects;
2. have HIV infection or have HIV-infected mothers;
3. critically-ill and cannot survive through the study period;
4. allergic to the study drugs.
