研究目的:

创伤后创面愈合并转变为瘢痕组织,其中增生性瘢痕不仅影响外观更影响瘢痕所在组织的生理功能。在已有的研究中已发现临床稳定的增生性瘢痕样本的成纤维细胞中二肽基肽酶4(Dpp4)与正常皮肤表达存在差异,本课题一方面希望进一步了解Dpp4是否通过TGF-β-Smad信号通路及相关基因甲基化程度的变化参与瘢痕表型的形成,从而了解Dpp4参与瘢痕形成的机制;另一方面通过观察阻断创伤愈合早期Dpp4表达是否关闭成纤维细胞的合成表型并最终抑制增生性瘢痕的形成,为瘢痕治疗提供新的方向。本实验设计通过抑制瘢痕成纤维细胞及兔耳瘢痕模型中Dpp4表达观察细胞凋亡及胶原沉积以及模型中瘢痕形成情况,以验证Dpp4在增生性瘢痕形成中的关键作用,并将进一步在细胞及动物模型中检测抑制Dpp4后相关信号通路各分子表达变化,及相关基因甲基化情况从而进一步筛选相关靶基因并在细胞及动物模型中加以验证。  

Objectives of Study:

Wound is healed with scar after severe injury almost in all patients. Some patients developed hypertrophic scar, while the hypertrophic scars not only affect the appearance but also the normal function. In our previous study, it has been found that the expression of dipeptidyl peptidase-4 (Dpp4) in fibroblasts form scar samples is different from that of normal skin. On one hand, the study is designed to further understand the function of Dpp4 in scar formation, and the involvement of the TGF-β-Smad signaling pathway and the degree of methylation of related genes participate in the formation of scar, thereby understanding the mechanism of Dpp4 involvement in scar formation. On the other hand, inhibiting Dpp4 in the early stage of wound healing by specific inhibitor to observe whether it could inhibit the synthesis of fibroblasts and ultimately inhibit the formation of hypertrophic scar, thus providing a new direction for scar treatment. This experiment designed to verify the key role of Dpp4 in the formation of hypertrophic scar by inhibiting the expression of Dpp4 in scar fibroblasts and rabbit ear model by observing apoptosis and collagen deposition in fibroblasts and scar formation in the model. Further, the expression of relevant molecule in TGF-β-Smad signaling pathway under inhibition of Dpp4, and the methylation status of related genes were detected to further screen related target genes. The methylation expression of related genes was inhibited and Dpp4 would be overexpressed in fibroblasts. The fibroblast would be observed for collagen deposition and apoptosis to verify the relationship between Dpp4, gene methylation and scar formation.

增生性瘢痕患者,年龄小于70周岁,无心脑血管及恶性肿瘤疾病

Inclusion criteria

Patients with hypertrophic scar, aged less than 70 years old, have no cardiovascular and malignant tumor diseases

排除标准:

患有严重心脑血管疾病,恶性肿瘤,怀孕妇女及12岁以下儿童

Exclusion criteria:

Suffering from serious cardiovascular and cerebrovascular diseases, malignant tumors, pregnant women and children under 12 years old
