本项目拟通过前期研究队列,筛选PM2. 5不同暴露水平下靶向RAGE的特异性miRNA,结合体内外实验,从分子、细胞、动物三个层面揭 示PM2.5不同暴露水平特异性miRNA与RAGE的表达变化及对炎症细胞和炎症介质的影响,探索特 异性miRNA的抑制或过表达对RAGE的调控作用与分子机制,阐明PM2.5在慢阻肺气道炎症中的作用,为寻找敏感的生物标志物及可能的药物干预靶点提供科学依据。 |

Objectives of Study: |
This study aims to screen specific miRNAs targeting RAGE at different exposure levels of PM2.5 through our previous research cohort. Combined with in vivo and in vitro experiments, we will reveal the expression changes of microRNA targeting RAGE at different exposure levels of PM2.5 and their effects on inflammatory cells and inflammatory mediators from molecule, cell and animal aspect, and explore the regulatory role and molecular mechanism of microRNAs in RAGE by functional inhibition or over expression. Ultimately, we hope to elucidate the role of PM2.5 in COPD airway inflammation and provide a scientific basis for finding sensitive biomarkers and possible drug intervention targets. |

①符合《慢性阻塞性肺疾病全球倡议》(GOLD-2020)诊断标准,入组时处于疾病稳定期; |

Inclusion criteria |
① It complies with the diagnostic criteria of the Global Initiative for Chronic Obstructive Pulmonary Disease (GOLD-2020)and in a stable disease period; |

排除标准: |
①有严重心脑血管疾病、肝肾功能不全、癫痫及其他神经精神性疾病、活动性肺结核等慢性疾病史或合并其他疾病影响信息收集; |

Exclusion criteria: |
1. people who have chronic diseases, such as severe cardiovascular and cerebrovascular diseases, liver or renal insufficiency, epilepsy and other neuropsychiatric diseases, active pulmonary tuberculosis, and the impact of other diseases or have other diseases influence the Information gathering; |
