研究目的：

1. 研究GCKR不同转录本的表达情况、蛋白编码能力及对应的蛋白功能； 2. 探索不同疾病状态（包括T2DM、脂肪肝、肝硬化、肝癌）下，肝脏组织中GCKR基因三种转录本的表达差异，探索其与疾病发生发展的关联； 3. 揭示GCKR不同转录本在调控肝脏糖脂代谢中的具体作用，并初步阐明其涉及的关键信号通路与分子机制，为后续靶向干预提供新策略。  

Objectives of Study：

1. Investigate the expression profiles, protein-coding potential, and corresponding protein functions of different GCKR transcripts. 2. Explore the expression differences of the three GCKR transcripts in liver tissues under various pathological conditions (including T2DM, fatty liver disease, cirrhosis, and hepatocellular carcinoma), and examine their associations with disease development and progression. 3. Elucidate the specific roles of different GCKR transcripts in regulating hepatic glucose and lipid metabolism, and preliminarily clarify the key signaling pathways and molecular mechanisms involved, thereby providing new strategies for targeted interventions.

纳入标准：

1: 因自身疾病拟于本中心接受肝脏部分切除手术的患者； 2: 无手术的医学或麻醉禁忌症； 3: 理解研究程序的性质，愿意参与并给予书面知情同意

Inclusion criteria

1: Patients scheduled to undergo partial hepatectomy at this center due to their own medical condition 2: No medical or anesthesia contraindications for surgery 3: Understand the nature of the study procedures, willing to participate, and provide written informed consent

排除标准：

1: 合并其他严重疾病：严重的肾功能不全，心血管疾病或严重疾病状态患者； 2: 术前接受过放疗、化疗、免疫或靶向治疗等； 3: 长期服用免疫抑制剂、激素类药物等； 4: 活动性病毒性肝炎；

Exclusion criteria：

1: Patients with other severe comorbidities: severe renal insufficiency, cardiovascular diseases, or other serious medical conditions 2: Patients who have received preoperative treatments such as radiotherapy, chemotherapy, immunotherapy, or targeted therapy 3: Long-term use of immunosuppressants, hormonal medications, etc. 4: Active viral hepatitis

数据采集和管理（说明：数据采集和管理由两部分组成，一为病例记录表(Case Record Form, CRF)，二为电子采集和管理系统(Electronic Data Capture, EDC)，如ResMan即为一种基于互联网的EDC：

数据采集：湘雅二医院电子病例系统收集病人病例资料和检测结果，由专人录入电子excel表后再由第二人检查数据有无错误。 数据管理：当所有数据经专人录入再由第二人检查无误后，将收集病例按入组顺序编号，病例资料按编号顺序保存于BIMS生物样本库管理系统，并设置多个备份保存于至少两个单独的存储盘上，防止损坏和丢失。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture：

Data Collection: Patient case records and test results are gathered through the Xiangya Second Hospital Electronic Medical Record System. A designated staff member enters the data into electronic Excel spreadsheets, which are then reviewed by a second person to verify accuracy. Data Management: After all data is entered by a designated staff member and verified by a second reviewer, collected cases are numbered in enrollment order. Case materials are stored in the BIMS Biobank Information Management System according to their assigned numbers. Multiple backups are maintained on at least two separate storage drives to prevent damage or loss.
