研究目的: |
1. 建立AAAD患者临床病历资料及生物学样本数据库:前瞻性收集AAAD患者的临床病历资料,采集患者的外周血单个核淋巴细胞(peripheral blood mononuclear cell,PBMC)及血浆样本,构建AAAD患者的临床病历资料及生物学样本数据库。 2. 揭示AAAD患者发生术后感染的分子机制:通过对AAAD患者的生物学样本进行多模态组学分析,包括:细胞因子芯片、免疫细胞图谱、单细胞测序、全转录组测序、蛋白组学分析及代谢组学分析,深入挖掘AAAD患者发生术后感染的分子机制。 3. 构建AAAD患者术后感染的预测模型:通过机器学习算法对患者的临床病历资料和生物学样本多模态组学测序结果进行整合和分析,构建AAAD患者术后感染的预测模型,并在临床患者中进行验证。 |

Objectives of Study: |
1. Establishment of a clinical medical record and biological sample database for AAAD patients: Prospectively collect clinical medical records of patients with acute ascending aortic dissection (AAAD), collect peripheral blood mononuclear cells (PBMC) and plasma samples, and construct a clinical medical record and biological sample database for AAAD patients. 2. Revealing the molecular mechanism of postoperative infection in AAAD patients: Perform multimodal omics analysis on biological samples of AAAD patients, including cytokine array, immune cell profiling, single-cell sequencing, whole-transcriptome sequencing, proteomic analysis, and metabolomic analysis, to deeply explore the molecular mechanism underlying postoperative infection in AAAD patients. 3. Construction of a prediction model for postoperative infection in AAAD patients: Integrate and analyze patients' clinical medical records and multimodal omics sequencing results through machine learning algorithms, construct a prediction model for postoperative infection in AAAD patients, and verify it in clinical patients. |

1. 患者术前经主动脉 CT 血管成像确诊为AAAD; 2. 患者住院期间在 CPB 下行外科手术治疗; 3. 患者签署知情同意书。 |

Inclusion criteria |
1. The patient was diagnosed with AAAD by preoperative aortic computed tomography angiography (CTA). 2. The patient underwent surgical treatment under cardiopulmonary bypass (CPB) during hospitalization. 3. The patient signed the informed consent form. |

排除标准: |
1. 术前合并感染,感染的部位及类别不限; 2. 术前存在严重的脏器功能不全,比如:大面积脑梗死、脑出血、肝功能衰竭、心肌梗死、心脏骤停、消化道大出血或严重的肾脏功能衰竭; 3. 住院期间未行外科手术,选择内科保守治疗的患者; 4. 患者同期参与其他的药物研究或临床试验; 5. 患者或其家属拒绝签定知情同意书; 6. 妊娠或哺乳期。 |

Exclusion criteria: |
1. Preoperative combined infection, regardless of the site or type of infection; 2. Severe organ dysfunction present before surgery, such as: large-area cerebral infarction, cerebral hemorrhage, liver failure, myocardial infarction, cardiac arrest, major gastrointestinal bleeding, or severe renal failure; 3. Patients who did not undergo surgery during hospitalization and chose conservative medical treatment; 4. Patients concurrently participating in other drug studies or clinical trials; 5. Patients or their family members refuse to sign the informed consent form; 6. Pregnancy or lactation. |
