总体目标: 阐明牙周炎关键生化标志物MMP-8的太赫兹波谱响应规律,突破现有牙周炎活动期诊断缺乏精准、快速技术的瓶颈,建立一种基于太赫兹分子指纹光谱的无创、椅旁诊断新方法,为牙周炎的精准防控提供全新的技术手段和理论依据。 具体目标: 解析规律:获得MMP-8在太赫兹波段的特征指纹图谱,明确其特征吸收峰位,并建立其浓度与太赫兹吸收强度的定量关系模型。 建立方法:优化并确立针对临床GCF样本的标准化太赫兹检测方案,实现在复杂生物基质中准确、稳定地检测MMP-8。 构建模型:基于太赫兹光谱特征,构建一个能够有效区分牙周炎活动期、静止期及健康状态的诊断模型,并初步验证其临床诊断效能(目标:AUC > 0.85)。 机制探索:从分子振动模式角度,初步探索太赫兹波与MMP-8蛋白相互作用的传感机制,为方法提供深层次科学解释。  

Objectives of Study:

Overall objectiveTo clarify the terahertz spectral response law of the key biochemical marker MMP-8 of periodontitis, break through the bottleneck of the lack of precise and rapid techniques for the diagnosis of active periodontitis in the existing system, and establish a new non-invasive and chair-based diagnostic method based on terahertz molecular fingerprint spectroscopy, providing a brand-new technical means and theoretical basis for the precise prevention and control of periodontitis.Specific goalsAnalysis rule: Obtain the characteristic fingerprint spectrum of MMP-8 in the terahertz band, clarify its characteristic absorption peak positions, and establish a quantitative relationship model between its concentration and terahertz absorption intensity.Establishment method: Optimize and establish a standardized terahertz detection protocol for clinical GCF samples to achieve accurate and stable detection of MMP-8 in complex biological matrices.Model construction: Based on terahertz spectral characteristics, a diagnostic model capable of effectively distinguishing between the active phase, stationary phase and healthy state of periodontitis was constructed, and its clinical diagnostic efficacy was preliminarily verified (objective: AUC > 0.85).Mechanism exploration: From the perspective of molecular vibration modes, the sensing mechanism of the interaction between terahertz waves and MMP-8 protein is preliminarily explored to provide a deep scientific explanation for

纳入标准:

试验组 1.自愿参与本研究; 2.有探诊深度>=6mm; 3.出血指数(bleeding onprobing, BOP)阳性的患牙. 对照组: 1.自愿参与本研究; 2.有牙周健康(探诊深度<=3mm,无炎症)且无龋坏的健康牙.

Inclusion criteria

Experimental group 1. Voluntarily participate in this study; 2. Have probing depth >= 6mm; 3. Teeth with positive bleeding on probing (BOP). Control group 1. Voluntarily participate in this study; 2. Healthy teeth with periodontal health (probing depth <= 3mm, no inflammation) and no caries.

排除标准:

试验组 1.近期(6 个月)经过牙周治疗; 2.有全身性疾病(如糖尿病等); 3.妊娠或哺乳期患者 对照组 1.有全身性疾病(如糖尿病等); 2.妊娠或哺乳期患者。

Exclusion criteria:
