研究目的：

该研究拟以膝骨关节炎患者和膝关节损伤患者为研究对象，通过脂质组学与转录组学联合分析，深入探讨膝骨关节炎发病进程中脂质代谢产物（脂肪酸、磷脂等）及相关调控基因的变化特征与作用机制，明确调控脂质代谢紊乱的关键基因靶点，并揭示其介导膝关节滑膜、软骨及脂肪垫损伤的分子机制，以期为膝骨关节炎发病机制提供新视角，为制定靶向脂质代谢的膝骨关节炎早期防诊治策略提供科学依据。  

Objectives of Study：

This study intends to take patients with knee osteoarthritis and patients with knee joint injury as the research objects. Through the combined analysis of lipidomics and transcriptomics, it will thoroughly explore the variation characteristics and mechanisms of action of lipid metabolites (such as fatty acids and phospholipids) and related regulatory genes during the pathogenesis of knee osteoarthritis, identify the key gene targets that regulate lipid metabolism disorders, and reveal the molecular mechanisms by which these targets mediate injuries to the knee joint synovium, cartilage, and fat pad. The purpose is to provide a new perspective for the pathogenesis of knee osteoarthritis and a scientific basis for formulating early prevention, diagnosis, and treatment strategies for knee osteoarthritis targeting lipid metabolism.

纳入标准：

1.病例组纳入标准： （1）儿童年龄6-17岁.成年人年龄大于18岁； （2）.符合《中国骨关节炎诊疗指南(2024版)》骨关节炎（OA）的临床诊断标准； （3）.拟行微创或开放手术治疗，术前未接受过脂质调节药物（如他汀类）长期治疗； （4）.自愿参与本研究并签署知情同意书。 2.对照组纳入标准： （1）儿童年龄6-17岁.成年人年龄大于18岁； （2）.无关节炎疾病病史，因膝关节损伤（如半月板损伤、韧带损伤等）拟行微创手术治疗； （3）.自愿参与本研究并签署知情同意书。

Inclusion criteria

1. Inclusion Criteria for the Case Group (1)Children are aged 6-17 years. Adults are older than 18 years; (2).Meeting the clinical diagnostic criteria for osteoarthritis (OA) as outlined in the Chinese Guidelines for the Diagnosis and Treatment of Osteoarthritis (2024 Edition); (3).Planned to undergo minimally invasive or open surgery, with no long-term prior administration of lipid-regulating medications such as statins before the operation; (4).Voluntarily participating in this study and signing the informed consent form. 2.Inclusion Criteria for the Control Group (1)Children are aged 6-17 years. Adults are older than 18 years; (2).No history of arthritis, and scheduled for minimally invasive surgery due to knee joint injuries such as meniscal lesions, ligament tears and other related conditions; (3).voluntarily participating in this study and signing the informed consent form.

排除标准：

1.排除标准： （1）.合并其他恶性肿瘤或严重肝肾功能不全者； （2）.近 3个月内接受过关节腔注射激素、非甾体抗炎药长期治疗或脂质调节药物治疗者； （3）.女性处于孕期、哺乳期或经期； （4）.拒绝提供完整临床资料或配合样本采集者。

Exclusion criteria：

1.Exclusion Criteria (1).Patients with comorbidities of other malignant tumors or severe hepatic and renal insufficiency; (2).Patients who have received intra-articular corticosteroid injections, long-term treatment with non-steroidal anti-inflammatory drugs, or treatment with lipid-regulating medications within the past 3 months; (3).Female patients who are in the pregnancy, lactation, or menstrual period; (4).Patients who refuse to provide complete clinical data or cooperate with sample collection.
